PF3D7_1315400 (PfMD3) is a cytoplasmic CCCH-type zinc finger RNA-binding protein expressed in immature male Plasmodium falciparum gametocytes that acts in post-transcriptional control to drive male gametocytogenesis and mosquito infectivity (PMID:38148574, PMID:42244567). It binds a defined set of mRNA transcripts encoding male gamete development proteins through a specific RNA sequence motif and promotes their translation: proteins encoded by PfMD3-bound transcripts fall sharply in knockout parasites, establishing a translation-promoting rather than purely stabilizing role (PMID:42244567). PfMD3 operates within an RNA-binding protein network that includes PABP1 and ALBA3, translational initiators/regulators/repressors (eIF4G, PUF1, NOT1, CITH), and the gametocytogenesis regulators ZNF4, MD1, and GD1 (PMID:38148574). Consistent with these interactions, targeted disruption of PfMD3 impairs gametocyte maturation and male gametocytogenesis and produces a male fertility defect with reduced infectivity to mosquitoes (PMID:38148574, PMID:42244567). Beyond this post-transcriptional regulatory role in male gamete development, no further mechanistic detail has been characterized in the available corpus.
