EXP3 (PF3D7_1024800) is a parasitophorous vacuole membrane-associated protein of Plasmodium that functions in the compartment where parasite effector proteins are staged for export into the host erythrocyte (PMID:37772444, PMID:33793667). In P. falciparum it is a component of the Exported Protein-Interacting Complex (EPIC), together with PV1 and PV2, and EPIC physically associates with the PTEX export translocon (including P113) (PMID:35403274); consistent with this, EXP3 is captured in proximity to the exported membrane protein SBP1 at an early, pre-export stage, placing it near cargo before translocation (PMID:37772444). Functionally, EXP3 behaves as an accessory rather than a core export factor: conditional depletion or disruption has little effect on bulk protein export or parasite fitness in vitro, while accessory factors of this class contribute to export of virulence-relevant effector subsets in vivo (PMID:33793667). The P. berghei ortholog (PBANKA_0509000) localizes to the PVM in both blood- and liver-stage parasites (PMID:29404413). Beyond its membership in EPIC and its PVM association, no direct enzymatic activity, structural model, or defined molecular mechanism for EXP3 itself has been characterized in the available corpus.
