UPF1 (PF3D7_1005500) is a cytoplasmic mRNA-associated protein of Plasmodium that, despite its annotation as the core NMD helicase, is dispensable for nonsense-mediated mRNA decay in P. falciparum blood-stage parasites (PMID:37366629). Epitope-tagging places PfUPF1 in cytoplasmic puncta, and reciprocal co-immunoprecipitation shows it physically associates with PfUPF2 and additional mRNA-binding proteins (PMID:37366629). CRISPR-Cas9 disruption neither stabilizes nonsense transcripts nor impairs asexual proliferation, establishing that PfUPF1 is non-essential for both NMD and blood-stage growth (PMID:37366629). Its association with translational-regulatory mRNP complexes is stage- and species-dependent: in P. berghei female gametocytes UPF1 is explicitly excluded from affinity-purified DOZI/CITH mRNPs (PMID:20169188), whereas in P. yoelii it becomes proximal to ALBA4 specifically in zygotes but not gametocytes (PMID:39777415). Beyond these findings, the enzymatic activity, RNA substrates, and mechanistic role of PF3D7_1005500 have not been characterized in the available corpus.
