Synthesis pass · prose summary of the discoveries below

RAP3 (PF3D7_0501500) is a rhoptry-resident protein of Plasmodium falciparum and the third component of the low-molecular-mass rhoptry complex it forms with RAP1 and RAP2 (PMID:12183575, PMID:10835342). It is the more recently duplicated paralog of RAP2, arising from a head-to-tail gene pair on chromosome 5, a duplication that postdates the divergence of Plasmodium species (PMID:12183575). Within the complex, RAP3 associates noncovalently with RAP1, and immunodepletion indicates that RAP3 and RAP2 bind one RAP1-class molecule in a mutually exclusive manner (PMID:1698657); this association forms early in the secretory pathway before rhoptry transport, and complex assembly requires intact RAP1 (PMID:10835342, PMID:9574915). RAP3 is dispensable for merozoite invasion and for rhoptry-mediated tagging of normal erythrocytes, functions that RAP2 can carry out in its absence (PMID:12183575, PMID:17624849). Conserved-region synthetic peptides of RAP3 directly bind red-blood-cell surface molecules of ~27–90 kDa and inhibit invasion in vitro, at binding sites shared or overlapping with those recognized by RAP1 and RAP2 peptides (PMID:20833215). In rodent malaria parasites, which encode a single RAP2/3 ortholog rather than distinct RAP2 and RAP3 genes, that ortholog localizes to rhoptries in merozoites and sporozoites, is secreted into the parasitophorous vacuole during invasion, and contributes to parasite virulence in vivo (PMID:19428669, PMID:24798694, PMID:29307564). No enzymatic activity or structural model for RAP3 has been characterized in the available corpus.

  1. 1990 Medium

Before its gene was cloned, biochemistry established that the p37 protein (RAP3) is a subunit of a defined rhoptry hetero-oligomer, framing RAP3 as a stoichiometric complex partner rather than a free protein.

Evidence Sucrose gradient centrifugation, crosslinking, and immunodepletion of merozoite extracts

    PMID:1698657

    Open questions at the time
    • RAP3 identity was inferred from the p37 product before gene cloning
    • Mutual exclusivity with RAP2 inferred from immunodepletion, not direct structure
    • No stoichiometry beyond minimal one-RAP1-plus-one
  2. 1998 Medium

The question of when in trafficking RAP3 joins the complex was addressed by showing association forms early in the secretory pathway before rhoptry delivery.

    PMID:12183575

    Open questions at the time
    • Redundancy with RAP2 inferred rather than directly shown by double knockout
    • No non-redundant function identified for RAP3
  5. 2007 Medium

A specific functional readout tested whether RAP3 mediates rhoptry tagging of erythrocytes, showing this function is carried by RAP1 and RAP2 and RAP3 is not needed.

Evidence Surface iodination of RAP3-disrupted mutant parasites

    PMID:17624849

    Open questions at the time
    • Negative result does not exclude a RAP3-specific role masked by RAP2
    • Molecular mechanism of tagging not addressed for RAP3
  6. 2010 Medium

Peptide biochemistry probed whether RAP3 engages the host cell directly, showing conserved RAP3 regions bind RBC surface molecules and inhibit invasion at sites shared with RAP1/RAP2 peptides.

Evidence Synthetic peptide RBC binding, competition, and in vitro invasion-inhibition assays

    PMID:20833215

    Open questions at the time
    • Peptide binding may not reflect native full-length protein behavior

2002 RAP3 (PF3D7_0501500) was identified as the third member of the low-molecular-mass rhoptry complex of P. falciparum. Disruption of the RAP3 gene demonstrated that RAP3 is not essential for merozoite invasion, likely because RAP2 can complement its loss. RAP3 shares homology with RAP2, and both genes are encoded on chromosome 5 in a head-to-tail arrangement. Phylogenetic analysis suggests the RAP2/RAP3 duplication occurred after divergence of Plasmodium species. 2000 RAP3 (PF3D7_0501500) forms part of the low-molecular-mass rhoptry complex together with RAP1 and RAP2. Immunoprecipitation experiments in RAP1-disrupted parasites showed that truncated RAP1 did not complex with RAP2 and RAP3, demonstrating that complex formation requires intact RAP1. 1990 RAP3 (identified as the p37 product) forms hetero-oligomeric complexes with RAP1 products (Pr86, p82, p67) via noncovalent interactions. Immunodepletion experiments suggested that p39 (RAP2) and p37 (RAP3) bind the RAP1 complex in a mutually exclusive manner, indicating the minimal complex composition is one RAP1-class molecule plus either RAP2 or RAP3. Sucrose gradient centrifugation, covalent crosslinking, immunoprecipitation, immunodepletion Experimental parasitology Medium 1698657
1998 RAP3 (p37 product) was detected in complexes with RAP1 precursor Pr86 and RAP2 (p39) in radiolabeled parasites treated with brefeldin A, suggesting complex formation occurs before rhoptry transport and that RAP3 associates with the RAP complex early in the secretory pathway. 2007 Using RAP3 gene-disruption mutant parasites and surface iodination experiments, RAP3 was shown NOT to be required for tagging of normal erythrocytes with rhoptry proteins (RAP1 and RAP2 carry this function). RAP3-disrupted parasites retained their capacity to tag erythrocytes, indicating RAP3 is dispensable for this process. 2010 Synthetic peptides derived from conserved regions of RAP3 (PF3D7_0501500) were shown to bind red blood cell (RBC) surface molecules of approximately 27–90 kDa in high-activity binding assays. These binding interactions were differentially sensitive to enzymatic treatments, and RAP3 peptides inhibited P. falciparum invasion in vitro. RAP1 and RAP2 peptides inhibited RAP3 peptide binding, suggesting recognition of overlapping or shared RBC binding sites. 2009 In P. berghei, GFP-tagged RAP2/3 (the single ortholog corresponding to RAP2 and RAP3) correctly targeted to merozoite rhoptries and also showed a distinct apical fluorescence pattern in sporozoites consistent with a rhoptry location, indicating expression and rhoptry localization of the RAP2/3 ortholog in two invasive zoite forms. 2014 In P. yoelii sporozoites expressing fluorescent RAP2/3, the tagged protein was secreted into the parasitophorous vacuole during host cell invasion (in contrast to RON2/RON4 which are lost upon rhoptry discharge), indicating a distinct secretory fate for the RAP2/3 protein during sporozoite invasion. Transgenic fluorescent protein tagging, live imaging of sporozoite invasion Cellular microbiology Medium 24798694
2018 In P. berghei, deletion of rap2/3 (the single ortholog) in the ANKA reference strain produced parasites with low parasitemia from which mice ultimately recovered. Complementation of RAP2/3 expression in the attenuated XAT strain partially restored virulence, demonstrating that RAP2/3 contributes to parasite virulence.
