Mechanistic narrative

Synthesis pass · prose summary of the discoveries below

PBANKA_1349200 (MSRP1) is an MSP7-like merozoite surface protein whose characterized biology centers on protein-protein interactions at the host-parasite interface and on transcriptional regulation during sexual commitment (PMID:12477792, PMID:26045295, PMID:41547993). MSRP1 was first identified as an interaction partner of the amino-terminal region of MSP-1, with its expression colocalizing with the MSP-1 N-terminus on the parasite surface (PMID:12477792). In P. berghei, MSRP1 directly binds host P-selectin through its N-terminus, engaging the C-type lectin and EGF-like domains of P-selectin; the resulting complex prevents P-selectin from binding its endogenous carbohydrate ligand Sialyl-Lewis(X), implicating MSRP1 in immunomodulation (PMID:26045295). In P. falciparum, MSRP1 expression is a downstream transcriptional target of the sexual commitment factor AP2-G: a DNA-binding-deficient AP2-G mutant abolishes MSRP1 expression, and MSRP1 expression marks early sexually committed schizonts (PMID:41547993). Genetic deletion of the P. berghei MSP7 family locus encompassing MSRP1 produces no detectable blood-stage or life-cycle phenotype, indicating MSRP1 is dispensable for asexual development in that species (PMID:26222913). Beyond these interaction, regulatory, and dispensability findings, no enzymatic activity or higher-resolution structural mechanism for MSRP1 has been characterized in the available corpus.

  1. 2002 Medium

Establishing whether MSRP1 physically associates with the merozoite surface complex, this work placed MSRP1 as a binding partner of MSP-1 rather than an independent surface protein.

Evidence Yeast two-hybrid screen, in vitro binding, and immunofluorescence colocalization with the MSP-1 N-terminus in P. yoelii and P. falciparum

2002 MSRP1 (designated MSRP-1 in P. falciparum) was identified as a protein that interacts with the amino-terminal region of MSP-1 in P. yoelii using the yeast two-hybrid system, and in vitro binding experiments confirmed the interaction between MSRP-1 and the amino-terminal region of P. falciparum MSP-1. Immunofluorescence studies colocalized MSRP expression with the amino-terminal portion of MSP-1 on the surfaces of trophozoites. 2015 In Plasmodium berghei, the ortholog of MSRP1 (referred to as PbMSRP1) was identified as a direct binding partner of host P-selectin. The interaction was mediated via the N-terminus of the MSP7-family protein and the C-type lectin and EGF-like domains of P-selectin. When complexed with PbMSRP1, P-selectin could no longer bind its endogenous carbohydrate ligand Sialyl-Lewis(X), suggesting a mechanism for immunomodulation. 2015 In P. berghei, a triple knockout of msp7/mspr1/mspr2 (the gene family including PBANKA_1349200/MSRP1) showed no impairment of life cycle progression, indicating that MSRP1 is dispensable for asexual blood-stage development in P. berghei. 2026 In P. falciparum, MSRP1 expression is dependent on the transcription factor AP2-G: parasites expressing a non-DNA-binding AP2-G.L2163 mutant did not express MSRP1, whereas functional AP2-G.V2163 was required for MSRP1 expression. This places MSRP1 as a downstream transcriptional target of AP2-G in the sexual commitment pathway, and its expression distinguishes early from late sexually committed schizonts. 2025 In P. falciparum, MSRP1 expression requires functional AP2-G (with valine at position 2163); a V2163L mutation that abolishes AP2-G DNA binding eliminates MSRP1 expression. MSRP1 expression can therefore be used as a marker to distinguish early and late sexually committed schizonts. This is a preprint version of the same finding as PMID:41547993. 2013 Members of the MSP-7 related protein (MSRP) family in P. falciparum, including MSRP members, were identified as PEXEL/HT-negative exported proteins (PNEPs), indicating unanticipated export into the host erythrocyte without a classical export motif. This was unexpected given the known surface localization of MSP7-family proteins. Reporter export assay, protein localization studies PLoS pathogens Low 23950716
