Mechanistic narrative

Synthesis pass · prose summary of the discoveries below

RON3 (PF3D7_1252100) is a rhoptry bulb membrane protein of Plasmodium falciparum merozoites that governs establishment of the intraerythrocytic ring stage after invasion (PMID:21237287, PMID:37457963). Unlike the moving-junction RON proteins, it partitions into a distinct RON complex, co-immunoprecipitating with PfRON2 and PfRON4 but not with PfAMA1 (PMID:21237287), and additionally associates with the rhoptry-associated membrane antigen RAMA in immature schizonts (PMID:33537242). Recombinant RON3 binds erythrocytes directly and anti-RON3 antibodies inhibit merozoite invasion, indicating a role at the host-cell interface (PMID:25495792). RON3 is proteolytically processed into N- and C-terminal fragments, and its C-terminal fragment is the functionally critical species: conditional disruption or stage-specific knockdown before invasion blocks erythrocyte invasion and arrests development beyond the ring stage, abolishes PTEX-dependent export of parasite proteins (e.g., RESA) across the parasitophorous vacuolar membrane, and abrogates glucose uptake into the parasitophorous vacuolar space while only partially affecting plasmodial surface anion channel activity (PMID:31289187, PMID:37457963). Knockdown after invasion does not perturb growth, placing RON3's essential action at or immediately before ring-stage establishment (PMID:37457963). Rhoptry localization is conserved in P. berghei sporozoites (PMID:31552198).

  1. 2011 Medium

Establishing where RON3 resides and which proteins it partners with was needed to distinguish it from the moving-junction machinery; localization to the rhoptry bulb and interaction with RON2/RON4 but not AMA1 defined RON3 as part of a separate RON complex that persists into the parasitophorous vacuole at the ring stage.

2011 RON3 (PF3D7_1252100) localizes to the rhoptry body (bulb), not the rhoptry neck, of P. falciparum merozoites, and remains detectable in the parasitophorous vacuole at the ring stage after invasion. Co-immunoprecipitation showed RON3 interacts with PfRON2 and PfRON4 but NOT with PfAMA1, indicating it is part of a distinct RON complex (RON2/3/4) separate from the moving junction complex. Immunofluorescence microscopy (IFA), immunoelectron microscopy, co-immunoprecipitation Parasitology international Medium 21237287
2014 Recombinant RON3 protein directly binds to erythrocytes in an erythrocyte-binding assay, and antibodies against RON3 significantly inhibit merozoite invasion into erythrocytes. Subcellular localization was confirmed by immunoelectron microscopy and IFA as the rhoptry bulb. 2019 Conditional disruption of RON3 in P. falciparum causes failure of exported proteins (e.g., RESA) to be transported across the PVM via PTEX, arrest of parasite development beyond the ring stage, and significantly decreased glucose uptake, establishing that RON3 influences both PTEX-dependent protein export and glucose transport into the parasitophorous vacuolar space. 2019 RON3 localizes to rhoptries in P. berghei sporozoites, as demonstrated by immuno-electron microscopy, showing that rhoptry localization of RON3 is conserved across both merozoite and sporozoite infective stages. 2021 RON3 forms a novel complex with rhoptry-associated membrane antigen (RAMA) in P. falciparum merozoites, identified by immunoprecipitation followed by LC-MS/MS. A region spanning amino acids Q221–E481 within RAMA may associate with RON3 in immature schizonts, though whether the interaction is direct or indirect remains unresolved. Immunoprecipitation, LC-MS/MS protein identification, immunofluorescence assay Frontiers in cellular and infection microbiology Medium 33537242
2023 RON3 is a rhoptry bulb membrane protein that undergoes proteolytic processing into N- and C-terminal fragments. Conditional knockdown of the RON3 C-terminal fragment from the early schizont stage impairs erythrocyte invasion and subsequent ring-stage development, abrogates protein export via PTEX and glucose uptake specifically at the late ring stage, while partially retaining plasmodial surface anion channel (PSAC) activity. Knockdown of the C-terminal fragment after invasion does not alter parasite growth, indicating its critical function is required before or at the time of invasion/early ring stage establishment.
