CITH (PF3D7_1474900) is a core structural component of a translationally repressive messenger ribonucleoprotein (mRNP)/stress granule complex in female Plasmodium gametocytes that sequesters maternally supplied mRNAs in a silent state prior to fertilization (PMID:20169188). Within this complex CITH is found together with the DEAD-box RNA helicase DOZI and integrates the cap-binding factor eIF4E and poly(A)-binding protein while excluding P body RNA-degradation factors and the initiation-promoting eIF4G, a composition consistent with storage rather than decay of transcripts (PMID:20169188). In P. berghei, CITH binds defined maternal transcript sets, including all nine cpw-wpc family mRNAs, holding them translationally silent until mosquito transmission (PMID:27312996), and in P. falciparum female gametocytes it localizes to stress granules and associates with DOZI, PABP1, and the seven-helix protein 7-Helix-1 (PMID:30133543). Genetic deletion of CITH yields fertilization-competent female gametocytes whose zygotes fail to develop into ookinetes, establishing CITH as a repressor essential for the transition to mosquito-stage development (PMID:20169188). Proximity proteomics across the gametocyte-to-zygote transition shows that this DOZI/CITH/ALBA repressive complex undergoes substantial compositional and spatial rearrangement upon fertilization—5' and 3' mRNA-associated proteins that are in proximity during repression dissociate in the zygote, coincident with a DOZI switch from NOT1-G to canonical NOT1—providing a trigger model for release of the stored transcripts (PMID:39777415).

2010 CITH (PF3D7_1474900 homolog in P. berghei) is a core component of a messenger ribonucleoprotein (mRNP) complex in female gametocytes, identified together with the RNA helicase DOZI by affinity purification coupled with mass spectrometry. This mRNP contains 16 major factors, integrates eIF4E and poly(A)-binding protein, excludes P body RNA degradation factors and translation-initiation-promoting eIF4G, and holds translationally silent transcripts. 2010 Gene deletion of CITH in P. berghei produces fertilization-competent gametocytes, but zygotes fail to develop into ookinetes in a female gametocyte-mutant manner. 2025 In P. yoelii, TurboID-based proximity proteomics of the DOZI/CITH/ALBA complex across the host-to-vector transmission event reveals that CITH is part of a translationally repressive complex in female gametocytes whose spatial arrangement and composition change substantially upon fertilization.
