Mechanistic narrative

Synthesis pass · prose summary of the discoveries below

PF3D7_1460400 (UCHL3) is an essential ubiquitin C-terminal hydrolase of Plasmodium falciparum that acts as a dual deubiquitinase and deNeddylase, removing both ubiquitin and Nedd8 modifications from substrates (PMID:17371404, PMID:20042598). Its catalytic behavior is governed by an active-site crossover loop that sterically restricts access of bulky adducts to the catalytic center; the loop is dispensable for catalysis per se, but enlarging it by mutation permits cleavage of substrates the wild-type enzyme cannot process, defining a size-restriction mechanism (PMID:19047059). Crystal structures of the apo enzyme and of a complex with the ubiquitin-based suicide substrate UbVME, together with modeling of the enzyme–Nedd8 complex, reveal the rigid crossover loop and the shared interactions that underlie dual ubiquitin/Nedd8 specificity, and the enzyme is essential for parasite survival (PMID:20042598). Functionally, inhibition of UCHL3 catalytic activity causes accumulation of polyubiquitinated proteins and disrupts blood-stage parasites, consistent with a role in ubiquitin removal from substrates (PMID:35042884), and its activity is required across multiple developmental stages including P. falciparum asexual blood stages and P. berghei sporozoites (PMID:37634761). The enzyme is druggable through distinct modalities: covalent fragments selective over human UCHL3/UCHL1 (PMID:37634761), constrained peptides that block the ubiquitin substrate-binding interface rather than the active site (PMID:38739798), and halofuginone, which binds UCHL3 directly and disrupts its influence on amino-acid biosynthetic protein expression (PMID:38923255). Beyond these findings, endogenous physiological substrates of UCHL3 in the parasite have not been defined in the available corpus.
