PF3D7_1453600 (RAP-1) is a rhoptry-associated protein of P. falciparum that forms the core of a multi-component antigen complex and functions post-invasion to support intraerythrocytic parasite growth (PMID:2200961, PMID:2263413, PMID:28205319). It is synthesized as a precursor carrying a cleavable N-terminal signal peptide and is trafficked to the rhoptries through the secretory pathway, its maturation from the 86-kDa precursor (Pr86) to mature p82 being reversibly blocked by brefeldin A and low temperature and irreversibly blocked by N-ethylmaleimide (PMID:8719244). Following transport to forming rhoptries, Pr86 undergoes progressive N-terminal cleavage to p82 and then p67, and assembles into low-molecular-weight complexes with the RAP-2 product p39 and the RAP-3 product p37 (PMID:9574915, PMID:17553576). RAP-1 is required for targeting RAP-2 to the rhoptries (PMID:29307564), and after merozoite release it binds the surface of normal erythrocytes, a tagging activity lost in RAP-1-disruption mutants (PMID:17624849). Rather than being required for rhoptry morphology or erythrocyte invasion, the RAP complex acts post-invasion to maintain normal parasitophorous vacuolar membrane structure and support intraerythrocytic growth, with conditional knockdown producing altered PVM, delayed growth, and reduced parasitemia (PMID:28205319). Monoclonal antibodies against RAP-1, including those recognizing inhibitory epitopes near the p80→p65 cleavage site and near the N-terminus of p67, inhibit parasite growth and invasion in vitro (PMID:2200961, PMID:2263413, PMID:1279419, PMID:9423886).

1990 RAP-1 is a rhoptry-associated protein of P. falciparum that forms a multi-component antigen complex consisting of four major proteins (80, 65, 42, and 40 kDa) and two minor proteins (77 and 70 kDa). Monoclonal antibodies against RAP-1 inhibit parasite growth in vitro. The antigen is lipophilic and disulfide bonding plays an important role in its structure. Autocatalytic processing and immunological cross-reactivity suggest all components are derived from the same precursor protein. 1992 Inhibitory monoclonal antibodies against RAP-1 recognize linear epitopes located 10–20 amino acids C-terminal to the proteolytic cleavage site (at amino acid 190) that converts the 80-kDa form (p80) to the 65-kDa form (p65). The 65-kDa product is absent in purified merozoites, suggesting its generation is associated with merozoite release rather than erythrocyte invasion. Epitope mapping using beta-galactosidase fusion proteins and synthetic peptides, Western blot, in vitro growth inhibition assay with purified immunoglobulin Molecular and biochemical parasitology High 1279419
1995 Transport of RAP-1 gene products to the rhoptries is regulated by the secretory pathway. Brefeldin A and low temperature reversibly block processing of the 86-kDa precursor (Pr86) to the mature 82-kDa RAP-1 product (p82). N-ethylmaleimide, which prevents vesicle–Golgi fusion, irreversibly blocks Pr86 processing. In vitro translation revealed a cleavable N-terminal signal peptide and an ~84-kDa primary translation product that differs from the in vivo precursor Pr86, suggesting an undetected protein modification occurs during maturation. Brefeldin A treatment, low-temperature block, N-ethylmaleimide treatment, in vitro translation, immunoprecipitation, metabolic labeling Molecular and biochemical parasitology High 8719244
1998 RAP-1 undergoes progressive N-terminal cleavage: the precursor Pr86 is converted to p82 and then to p67 by sequential removal of N-terminal sequences. Pr86 forms complexes containing RAP-2 product p39 and RAP-3 product p37. Processing of Pr86 begins after complexes are transported to forming rhoptries. Pr86 is present in rhoptries but not in segmenting schizonts or extracellular merozoites; p67 and p82 are major RAP-1 components in purified merozoites. Neither p67 nor p82 relocalize from the rhoptries to the merozoite surface under conditions that relocalize AMA-1. Immunoblot with epitope-specific antibodies, immunofluorescence, size fractionation, metabolic radiolabeling with brefeldin A, purified merozoite analysis Molecular and biochemical parasitology High 9574915
1998 A second inhibitory B-cell epitope (iB-2) near the amino terminus of p67 was identified in RAP-1. Monoclonal antibody 1D6 binds iB-2 and inhibits parasite invasion of erythrocytes in vitro. iB-2 is distinct from the previously identified iB-1 epitope (N200–Y211). Both inhibitory epitopes are located near the amino terminus of p67, a RAP-1 processing product implicated in merozoite invasion. 2007 RAP-1 is the core component of two low-molecular-weight rhoptry complexes (RAP1/RAP2 and RAP1/RAP3) identified in detergent-resistant membranes (DRMs) of P. falciparum schizonts. RAP-1 predominates in high-molecular-weight species by BN-PAGE, suggesting aggregation into larger complexes. RAP-1 was the most abundant protein detected by mass spectrometry in DRM fractions. Detergent-resistant membrane purification, blue native PAGE (BN-PAGE), mass spectrometry Molecular and biochemical parasitology Medium 17553576
2007 RAP-1 and RAP-2 (but not RAP-3) bind to the surface of normal (non-parasitized) erythrocytes. Surface iodination experiments showed that RAP-1 mutant parasites lose their capacity to tag erythrocytes, implicating RAP-1 specifically in erythrocyte tagging after merozoite release. RAP-1 gene-disruption mutants, surface iodination, erythrocyte binding assay The Journal of infectious diseases High 17624849
2017 The RAP complex (including RAP-1) is not required for rhoptry morphology or erythrocyte invasion, but is required post-invasion for normal parasitophorous vacuolar membrane (PVM) structure and intraerythrocytic parasite growth. Conditional knockdown of RAP complex members leads to altered PVM structure, delayed intraerythrocytic growth, and reduced parasitemia in infected mice. Conditional knockdown, electron microscopy (PVM morphology), growth assays in vitro and in vivo (mouse model) Cellular microbiology High 28205319
2018 In P. berghei, RAP-1 is required for targeting of RAP-2 to the rhoptries. RAP1-deficient parasites showed increased parasitemia early in infection but mice survived longer, indicating RAP-1 contributes to parasite virulence. RAP2/3-deficient parasites showed low parasitemia and full host recovery. Comparative genomics, RAP1-deficient and RAP2/3-deficient mutant generation, mouse infection assays Experimental parasitology Medium 29307564
1992 Three matrix metalloproteinases (220, 95, and 70 kDa) co-isolate with the RAP-1 complex by non-covalent association. These enzymes are inhibited by EDTA (indicating metal dependence) and enhanced by cobalt. Immunoprecipitation with gelatinase-specific antisera confirmed the metalloproteinases co-purify with RAP-1, but these antisera did not react with intracellular parasites by IFA, indicating the metalloproteinases originate from human serum used in culture, not from the parasite itself. Co-purification, immunoprecipitation, proteinase inhibitor panel testing, indirect immunofluorescence Biochimica et biophysica acta Low 1477099
2010 RAP-1 was identified as a component of a large MSP-1-associated surface protein complex in P. falciparum merozoites. Pull-down experiments with anti-MSP-1 or anti-RhopH3 antibodies from merozoite lysate co-precipitated RAP-1 and RAP-2 alongside MSP-3, MSP-6, MSP-7, MSP-9, RhopH1, RhopH3, and two RAP domain-containing proteins. This complex was detected at schizont/merozoite stages but not in ring stages. Antibody pull-down from merozoite lysate, tandem mass spectrometry Journal of proteome research Low 21175202
