PF3D7_1408200 (AP2-G2) is an AP2-domain transcription factor that acts as a global transcriptional repressor operating within the ApiAP2 regulatory cascade that governs the transition from asexual proliferation to sexual development in Plasmodium (PMID:26417110, PMID:28081440, PMID:33368818). It sits downstream of the master commitment regulator AP2-G: single-cell transcriptomics in P. falciparum show AP2-G2 is upregulated in sexually committed cells (PMID:29094698), and AP2-G directly binds upstream of AP2-G2 in P. berghei (PMID:34119684). Genome-wide ChIP-seq establishes that AP2-G2 binds ~1,500 gene promoters through a five-base cis-acting negative regulatory motif and that most targets encode asexual-proliferation functions, so its loss de-represses the asexual program without preventing sexual commitment (PMID:26417110); in P. falciparum it additionally binds within the gene bodies of ~3,000 genes in correlation with H3K36me3 and HP1 occupancy, and its disruption causes overexpression, but no downregulation, of bound genes, confirming repressor activity (PMID:33368818). Functionally, AP2-G2 is required for gametocyte maturation past stage III, and its disruption arrests sexual parasites before sex-specific morphologies emerge (PMID:26417110, PMID:33368818), with a disproportionate loss of male gametocytes traceable to direct activation of the male development gene PfMDV-1, whose transcription start site AP2-G2 occupies (PMID:33537025). AP2-G2 localizes to the nucleus (PMID:33537025) and physically associates with chromatin remodeling proteins, a MORC protein, and the ApiAP2 factor PF3D7_1139300 (PMID:33368818), and is found in a gametocyte-specific SAGA-like chromatin complex linked to a defined histone code (PMID:35051657), indicating it exerts repression in the context of chromatin-modifying machinery.

2014 AP2-G2 (PBANKA_103430) gene deletion in P. berghei significantly modulates but does not abolish gametocytogenesis, indicating it plays a modulatory role in commitment to gametocyte production and maturation, downstream of AP2-G in a cascade of ApiAP2 proteins. 2015 AP2-G2 functions as a transcriptional repressor in P. berghei gametocytes. Disruption of AP2-G2 did not prevent commitment to the sexual stage but halted development before sex-specific morphologies appeared. ChIP-seq revealed AP2-G2 targets ~1,500 genes by recognizing a five-base promoter motif; most target genes are required for asexual proliferation. Microarray analysis showed ~70% of up-regulated genes in AP2-G2-depleted parasites were AP2-G2 targets. A promoter assay demonstrated the AP2-G2 binding motif functions as a cis-acting negative regulatory element. 2017 AP2-G2 was identified as a repressor active in both asexual and sexual stages of P. berghei. A systematic knockout screen across ApiAP2 genes revealed AP2-G2 as a transcriptional repressor, and comparative transcriptomics across mutants and developmental stages revealed clusters of co-regulated genes with shared cis promoter elements whose expression is controlled negatively by AP2-G2. 2017 Single-cell RNA-seq of sexually committed AP2-G+ mature schizonts in P. falciparum revealed upregulation of additional AP2 transcription factors (including AP2-G2/PF3D7_1408200) in sexually committed cells, placing AP2-G2 as a downstream target of AP2-G during the commitment cycle. 2021 In P. falciparum, PfAP2-G2 (PF3D7_1408200) plays a critical role in gametocyte maturation. PfAP2-G2 binds promoters of a wide array of genes across parasite life cycle stages and also binds within gene bodies of ~3,000 genes, which correlates with H3K36me3, other histone modifications, and HP1 occupancy, suggesting gene body binding as an alternative regulatory mechanism. Disruption of pfap2-g2 does not impact asexual development but causes the majority of sexual parasites to arrest at stage III gametocytes. Absence of pfap2-g2 leads to overexpression of 28% of bound genes with no downregulation of bound genes, confirming repressor function. PfAP2-G2 co-immunoprecipitates with chromatin remodeling proteins, a MORC protein, and another ApiAP2 protein (PF3D7_1139300). 2021 AP2-G2 is identified as a target gene directly bound by AP2-G in P. berghei, placing AP2-G2 downstream in a transcriptional cascade initiated by AP2-G that promotes gametocyte development; AP2-G binding sites exist upstream of AP2-G2 in the genome. 2021 In P. falciparum, PfAP2-G2 (PF3D7_1408200) knockout leads to ~95% decrease in mature gametocyte numbers with a greater impact on male gametocyte production, resulting in a higher female/male ratio. RNA-seq and RT-qPCR showed disruption of PfAP2-G2 causes downregulation of PfMDV-1 (male development gene-1, PF3D7_1216500). ChIP-qPCR demonstrated PfAP2-G2 is enriched at the transcriptional start site of PfMDV-1 in both ring and schizont stages. PfAP2-G (the master regulator) is also downregulated in pfap2-g2 knockout schizonts but not in ring stages. 2021 Indirect immunofluorescence assay in P. falciparum showed nuclear localization of PfAP2-G2 in asexual blood stages, consistent with its role as a transcription factor. Indirect immunofluorescence assay (IFA) Frontiers in microbiology Medium 33537025
2021 In P. falciparum mature gametocytes, a gametocyte-specific SAGA-like effector complex including PfAP2-G2 was identified by chromatin proteomics and tied to the specific histone code H3R17me2K18acK23ac, suggesting PfAP2-G2 participates in a chromatin regulatory complex associated with this histone modification in mature gametocytes.
