Synthesis pass · prose summary of the discoveries below

Pcalp (PF3D7_1362400) is an essential calcium-dependent cysteine protease that governs intraerythrocytic cell cycle progression in Plasmodium falciparum (PMID:19164769). Genetic evidence establishes both the gene and its proteolytic activity as required: gene disruption and catalytic-cysteine allelic replacement both fail, and conditional knockdown delays the ring-to-trophozoite transition and blocks S-phase entry (PMID:19164769). The enzyme is active as a monomer through its catalytic subdomain IIa, which carries a Cys-His-Asn catalytic triad whose mutation abolishes protease activity in vitro (PMID:23374507), and in live trophozoites its activity is calcium-dependent and sensitive to the cysteine-protease inhibitor E-64 and the calpain-selective inhibitor ALLN (PMID:25281458). Pcalp is targeted to the nucleolus through an N-terminal lipidation program — myristoylation at Gly2 and palmitoylation at Cys3 and Cys22 — with palmitoylation status dictating nucleolar versus nuclear localization (PMID:19239622). Beyond a possible interaction with Pf-Hsp60 (PMID:26797432), the physiological substrates through which Pcalp drives cell cycle progression have not been characterized in the available corpus.

Mechanistic history

Synthesis pass · year-by-year structured walk · 7 steps
  1. 2003 Low

Before any functional work, it was unknown whether P. falciparum encoded a calpain at all; genome analysis established a single calpain homolog and framed its predicted enzymatic identity and candidate substrates.

Evidence bioinformatic genome analysis, PEST-score prediction, and BLAST homology searches

    PMID:12857354

    Open questions at the time
    • purely computational, no experimental validation of catalytic activity or substrates
    • absence of a calpastatin homolog leaves the regulatory mechanism unaddressed
  2. 2009 High

Having a candidate gene, the question became where the protein acts and how it gets there; endogenous tagging and lipidation-site mutagenesis showed nucleolar targeting controlled by N-terminal myristoylation and palmitoylation.

Evidence C-terminally endogenously tagged Pcalp, myristoylation/palmitoylation site mutagenesis, and chimeric reporters imaged in parasite and mammalian cells

    PMID:19239622

    Open questions at the time
    • the enzymes catalyzing myristoylation and palmitoylation are not identified
    • no functional consequence of nucleolar versus nuclear localization for catalytic activity established
  3. 2009 Low

A complementary antibody-based study addressed stage-specific expression, reporting cytoplasmic signal across erythrocytic stages that rises from late ring into early trophozoite.

Evidence polyclonal antibodies against Pcalp-specific peptides and confocal immunofluorescence across erythrocytic stages

    PMID:19861123

    Open questions at the time
    • cytoplasmic localization contrasts with the endogenously tagged nucleolar signal, and the discrepancy is unresolved
    • no functional consequence linked to the expression pattern
  4. 2009 High

The central question of biological importance was answered by genetics: multiple failed knockouts, a failed catalytic-Cys replacement, and conditional knockdown showed Pcalp and its proteolytic activity are essential and required for cell cycle progression.

Evidence attempted single/double cross-over disruption, catalytic-Cys missense allelic replacement, FKBP-Shield1 conditional knockdown with growth and cell-cycle staging

    PMID:19164769

    Open questions at the time
    • the molecular substrates whose cleavage drives the ring-to-trophozoite and S-phase transitions are unidentified
    • how essentiality relates to nucleolar localization is not established
  5. 2013 Medium

The architecture of the active enzyme was unresolved; recombinant work showed the active form is catalytic subdomain IIa alone, functioning as a monomer via a Cys-His-Asn triad.

Evidence heterologous expression of recombinant subdomain IIa, fluorogenic substrate assay, gelatin zymography, catalytic-triad mutagenesis, and homology modeling

    PMID:23374507

    Open questions at the time
    • single-lab in vitro finding without full-length enzyme comparison
    • no structural determination beyond homology modeling
    • substrate specificity of the isolated subdomain not defined
  6. 2014 Medium

Whether the protease behaves as a calcium-dependent calpain inside the living parasite was tested directly, confirming calcium-dependent cysteine-protease activity in intraerythrocytic trophozoites.

Evidence live-cell fluorogenic substrate (Z-Phe-Arg-MCA) assays with E-64 and ALLN inhibition and Ca2+ modulation by calmidazolium and thapsigargin

    PMID:25281458

    Open questions at the time
    • the fluorogenic substrate is not Pcalp-specific, so activity attribution is inferential
    • the source and dynamics of the activating Ca2+ signal are not resolved
  7. 2015 Low

Two studies probed pharmacological and physical context: organotelluranes were identified as inhibitors of the calcium-stimulated protease, and a candidate Hsp60 partnership was reported.

Evidence intracellular protease inhibition assays with Ca2+-ATPase discrimination and growth assays (3D7, W2); co-immunoprecipitation, immunofluorescence co-localization, and mizoribine treatment for the Hsp60 link

    PMID:26384965 PMID:26797432

    Open questions at the time
    • organotellurane targeting is inferred without direct binding to purified Pcalp
    • Hsp60 interaction rests on a single non-reciprocal Co-IP and a non-specific inhibitor
    • functional consequence of the putative Hsp60 association is undefined

Open questions

Synthesis pass · forward-looking unresolved questions
  • The identity of the physiological substrates cleaved by Pcalp to drive the ring-to-trophozoite transition and S-phase entry, and how its nucleolar localization couples to this cell-cycle role, remain unknown.
  • no validated endogenous substrate identified
  • regulation in the absence of a calpastatin homolog is unexplained
  • reconciliation of nucleolar versus cytoplasmic localization reports is unresolved

2009 Pcalp (PF3D7_1362400) localizes to the nucleolus of P. falciparum parasites. The protein is myristoylated at Gly2 and palmitoylated at Cys3 and Cys22 in its N-terminal domain. Palmitoylation status dictates nucleolar vs. nuclear localization. An embedded nuclear localization sequence confers nucleolar targeting. These targeting signals function in mammalian cells as well as in the parasite. 2009 Pcalp (PF3D7_1362400) is essential for P. falciparum intraerythrocytic development. Gene disruption (double cross-over and single cross-over) and allelic replacement with a catalytic-cysteine missense mutation all failed, while synonymous allelic replacement succeeded, indicating that both the gene and its proteolytic activity are required. Conditional knockdown via FKBP-Shield1 system showed that depletion of Pcalp greatly impairs parasite growth and causes a delay in transition out of ring stage and failure to progress to S phase, placing Pcalp in cell cycle progression. 2013 The active form of Pcalp (PF3D7_1362400) consists solely of catalytic subdomain IIa, which contains the catalytic triad (Cys-His-Asn) and functions as a monomer. Recombinant subdomain IIa displayed protease activity measured by fluorogenic substrate assay and gelatin zymography; mutation of catalytic triad residues abolished activity. 2014 Pcalp (PF3D7_1362400) displays intracellular calcium-dependent cysteine protease activity detectable in live intraerythrocytic trophozoite-stage P. falciparum. Activity was measured using the fluorogenic substrate Z-Phe-Arg-MCA and was inhibited by the cysteine protease inhibitor E-64 and by the calpain-selective inhibitor ALLN. Manipulation of cytosolic Ca2+ via calmodulin inhibitor (calmidazolium) or SERCA inhibitor (thapsigargin) modulated Pcalp activity, establishing calcium dependence in the parasite cell.
