Synthesis pass · prose summary of the discoveries below

PK5 (PfPK5) is a cyclin-dependent kinase homologue of Plasmodium falciparum that functions in cell-cycle regulation during the intra-erythrocytic asexual stage, where it is implicated in the control of S-phase DNA synthesis (PMID:8143724, PMID:8943149). Recombinant PfPK5 is an intrinsically active kinase that phosphorylates histone H1 and casein and is present at approximately constant cytosolic levels across asexual development (PMID:8143724). Its activity is stimulated in vitro by cyclins — including the parasite cyclin homologue Pfcyc-1 and additional cyclin-related proteins (Pfcyc-2/3/4), the non-cyclin activator RINGO, and heterologous human cyclin H and p25 — and PfPK5 autophosphorylates in the presence of a cyclin, yet it is neither activated nor phosphorylated by the parasite CDK-activating kinase Pfmrk (PMID:10722743, PMID:12869562). PfPK5 is negatively regulated by CDK-inhibitor-type proteins, being inhibited in vitro by human p21(CIP1) but not by the CDK4/6-specific p16(INK4) (PMID:11700040). A direct substrate is the origin recognition complex subunit PfORC1: PfPK5-mediated phosphorylation of the ORC1 N-terminal domain abolishes its DNA-binding activity, alters its subcellular localization, and triggers proteasome-mediated degradation of ORC1 in schizonts, coupling PfPK5 activity to control of DNA replication licensing (PMID:26094711). The crystal structure of PfPK5, the first for a P. falciparum kinase, reveals a conserved monomeric CDK fold with inhibitor-binding-site differences from human CDKs that permit selective ATP-competitive inhibitor design (PMID:14604523).

1994 PK5 (PfPK5) expressed in E. coli as a recombinant protein displays kinase activity against casein and histone H1 substrates. Western blotting with polyclonal antibody showed the kinase is located in the parasite cytosol and is present in approximately constant amounts throughout the intra-erythrocytic asexual reproductive stage. 1994 PK5 (PfPK5) failed to complement cdc2/cdc28 yeast mutants, though the gene was not expressed in yeast, leaving open the question of functional equivalence to cdc2. Yeast complementation assay European journal of biochemistry Low 8143724
1996 PK5 (PfPK5) immunoprecipitated from aphidicolin-blocked parasites shows elevated kinase activity, suggesting involvement in S-phase regulation. Treatment of parasite cultures with CDK inhibitors flavopiridol and olomoucine (which inhibit PfPK5 in vitro at concentrations similar to other CDKs) markedly inhibited DNA synthesis during nuclear division cycles. Immunolocalization provided further evidence for a role in S-phase. 2000 PK5 (PfPK5) is efficiently activated in vitro by heterologous human cyclin H and p25 (a cyclin-like molecule that activates human CDK5), but is neither activated nor phosphorylated by Pfmrk (the putative P. falciparum CDK-activating kinase). PfPK5 can autophosphorylate in the presence of a cyclin. A novel P. falciparum cyclin homologue, Pfcyc-1, activates recombinant PfPK5 in vitro with efficiency similar to human cyclin H and p25. Activated PfPK5 can be inhibited by chemical CDK inhibitors. 2001 PK5 (PfPK5) is inhibited by heterologous human p21(CIP1) in vitro, but is not inhibited by the CDK4/6-specific inhibitor p16(INK4), suggesting that plasmodial CDKs may be regulated by inhibitory proteins in vivo in a manner partially conserved with mammalian CDKs. 2003 Three novel P. falciparum cyclin-related proteins (Pfcyc-2, Pfcyc-3, Pfcyc-4) activate PfPK5 in vitro, as evidenced by their ability to stimulate histone H1 kinase activity in association with PfPK5. Additionally, RINGO (a non-cyclin CDK activator) also strongly activates PfPK5 in vitro. In vitro kinase activation assay with recombinant proteins, histone H1 kinase assay The Journal of biological chemistry Medium 12869562
2003 The crystal structure of PK5 (PfPK5) was determined, representing the first structure of a P. falciparum protein kinase and the first CDK structure from a non-human source. The fold and mechanism of inactivation of monomeric CDKs are highly conserved. PfPK5 was co-crystallized with three ATP-competitive CDK inhibitors, revealing sequence differences from human CDKs within inhibitor binding sites that suggest selective inhibition is achievable. X-ray crystallography, co-crystallization with inhibitors Structure (London, England : 1993) High 14604523
2015 PK5 (PfPK5) phosphorylates the N-terminal domain of PfORC1 (Origin Recognition Complex subunit 1) in vitro using recombinant proteins. This phosphorylation abolishes DNA-binding activity of PfORC1, leads to changes in its subcellular localization, and triggers proteasome-mediated degradation of PfORC1 in schizonts, as demonstrated using transgenic parasites expressing wild-type or phosphorylation-mutant forms of ORC1. In vitro kinase assay with recombinant proteins, transgenic parasite expression of WT and mutant ORC1, subcellular localization analysis, proteasome inhibitor experiments Molecular microbiology High 26094711
1998 The CRK2 proteins of P. knowlesi and P. berghei are highly homologous to P. falciparum PfPK5. However, unlike PfPK5, a recombinant monomeric P. knowlesi CRK2 protein showed no kinase activity, and neither the P. knowlesi nor P. berghei CRK2 recombinant protein was able to complement a yeast cdc28ts mutant.
