Synthesis pass · prose summary of the discoveries below

PF3D7_1355200 (RAP-1) is a rhoptry bulb protein of Plasmodium falciparum that serves as the organizing core of the low-molecular-weight rhoptry protein complexes and, following invasion, contributes to parasitophorous vacuolar membrane (PVM) integrity and intraerythrocytic growth (PMID:28205319). It is synthesized as an ~84–86 kDa precursor bearing a cleavable N-terminal signal peptide and is trafficked to the rhoptries through the classical secretory pathway, since processing of the 86-kDa precursor to mature p82 is reversibly blocked by brefeldin A and low temperature and irreversibly blocked by N-ethylmaleimide (PMID:8719244). Within the rhoptries the precursor is proteolytically matured in sequential steps — rapid cleavage to p82 and delayed cleavage to p67 — each removing N-terminal epitopes, with a cleavage site near residue 190 and inhibitory epitopes clustered just C-terminal to it (PMID:1279419, PMID:9574915). RAP-1 forms the core of two distinct complexes, RAP-1/RAP-2 and RAP-1/RAP-3, resolved in detergent-resistant membranes of schizonts (PMID:17553576, PMID:9574915), and it is required for correct targeting of RAP-2 to the rhoptries (PMID:29307564). Contrary to a role in invasion itself, conditional knockdown of RAP complex members leaves rhoptry morphology and invasion intact but alters PVM structure and delays intraerythrocytic growth, defining a post-invasion function (PMID:28205319). RAP-1 also directly mediates binding to the surface of non-parasitized erythrocytes, an activity lost upon gene disruption (PMID:17624849). In P. berghei, deletion of RAP-1 modulates parasitemia kinetics and host survival, linking it to virulence (PMID:29307564).

1990 RAP-1 is a rhoptry-associated protein consisting of multiple components (80, 65, 42, 40, 77, and 70 kDa proteins) that are immunologically cross-reactive and appear to be derived from the same precursor protein; the antigen is lipophilic and disulfide bonding plays an important role in its structure. Immunoaffinity chromatography, SDS-PAGE, Western blot, autocatalytic processing analysis, monoclonal antibody epitope mapping Molecular and biochemical parasitology Medium 2200961
1992 RAP-1 is processed from an 80-kDa precursor (p80) in early schizogony to a 65-kDa form (p65) in late schizogony; inhibitory monoclonal antibodies map their epitopes to a region 10–20 amino acids C-terminal to the proteolytic cleavage site at amino acid 190; the p65 form is absent from purified merozoites, suggesting its generation is associated with merozoite release rather than erythrocyte invasion. Western blot, beta-galactosidase fusion protein epitope mapping, synthetic peptide reactivity assays, in vitro growth inhibition assays Molecular and biochemical parasitology Medium 1279419
1995 Transport of the RAP-1 gene product to the rhoptries is regulated by the secretory pathway: brefeldin A and low temperature reversibly block processing of the 86-kDa precursor (Pr86) to the mature 82-kDa RAP-1 (p82); N-ethylmaleimide (which prevents Golgi vesicle fusion) irreversibly blocks this processing. In vitro translation revealed a cleavable N-terminal signal peptide and an 84-kDa primary translation product, indicating the RAP-1 product acquires an additional protein modification during maturation. Brefeldin A and low-temperature block experiments on intact cells, N-ethylmaleimide treatment, in vitro translation, immunoblot analysis Molecular and biochemical parasitology High 8719244
1998 RAP-1 precursor Pr86 undergoes two sequential processing events in the rhoptries: rapid cleavage to p82 and delayed cleavage to p67, both involving progressive removal of epitopes from the amino-terminus. Pr86 forms complexes that also contain RAP-2 product p39 and RAP-3 product p37. p67 and p82 are the major RAP-1 forms in purified merozoites, but are not relocalized to the merozoite surface under conditions that promote AMA-1 relocalization. Since p67 is absent from ring-stage parasites, it may function during a narrow window of merozoite invasion. Immunoblot with epitope-mapped antibodies, size fractionation with brefeldin A block, immunofluorescence localization, merozoite purification Molecular and biochemical parasitology High 9574915
1998 Two inhibitory epitopes (iB-1 at N200–Y211, and iB-2) in RAP-1 are located near the amino-terminus of p67. Anti-iB-1 antibodies are produced during infection but are not immunodominant and are not readily boosted upon reinfection in owl monkeys. A monoclonal antibody (1D6) binding iB-2 inhibits erythrocyte invasion in vitro. Owl monkey immunization and reinfection, ELISA antibody mapping, in vitro invasion inhibition assay with monoclonal antibodies Infection and immunity Medium 9423886
2007 RAP-1 is a core component of two low molecular weight rhoptry complexes (RAP1/RAP2 and RAP1/RAP3) identified in detergent-resistant membranes (DRMs) of schizonts by blue native-PAGE and mass spectrometry; RAP-1 is highly abundant in high-molecular-weight DRM species, suggesting participation in larger aggregated complexes. DRM purification, blue native-PAGE, mass spectrometry Molecular and biochemical parasitology Medium 17553576
2007 RAP-1 and RAP-2 (but not RAP-3) bind to the surface of normal (non-parasitized) erythrocytes; RAP-1 gene-disruption mutants lose the capacity to tag bystander erythrocytes, implicating RAP-1 directly in this binding activity. RAP-1 gene-disruption mutant parasites, surface iodination experiments, antibody binding assays on normal erythrocytes The Journal of infectious diseases High 17624849
2010 PfRAP-1 is identified as part of a large merozoite surface protein-1 (MSP-1) associated complex: pull-down experiments with anti-MSP-1 or anti-RhopH3 antibodies co-isolated PfRAP-1 and PfRAP-2, along with MSP-3, MSP-6, MSP-7, MSP-9, RhopH1, and RhopH3. The complex is present at schizont/merozoite stages and shed into culture supernatant. Co-immunoprecipitation/pull-down with merozoite lysate, tandem mass spectrometry identification Journal of proteome research Low 21175202
2017 The RAP complex (including RAP-1) is not required for rhoptry morphology or erythrocyte invasion per se; instead, conditional knockdown of RAP complex members leads to altered parasitophorous vacuolar membrane (PVM) structure, delayed intraerythrocytic growth, and reduced parasitemia in infected mice, indicating a post-invasion role in PVM integrity and parasite survival within the host cell. Conditional knockdown of RAP complex members (P. falciparum and P. berghei), electron microscopy of PVM, parasitemia measurements in mice Cellular microbiology High 28205319
2018 In P. berghei, RAP-1 is required for targeting of RAP-2 to the rhoptries; deletion of RAP-1 in P. berghei ANKA resulted in increased early-phase parasitemia but prolonged host survival compared to controls, indicating RAP-1 contributes to parasite virulence. Genetic deletion of RAP-1 in P. berghei ANKA, immunofluorescence localization of RAP-2, mouse infection outcome monitoring Experimental parasitology Medium 29307564
1992 Three matrix metalloproteinases (220, 95, and 70 kDa) co-isolate with the RAP-1 complex by immunoaffinity chromatography, but these enzymes originate from human serum used to culture P. falciparum in vitro (not from the parasite itself), as confirmed by immunoprecipitation with macrophage/fibroblast gelatinase antisera that did not react with intracellular parasites by IFA. Co-immunoaffinity isolation, proteinase inhibitor panel, metal ion dose-response, immunoprecipitation with anti-gelatinase antisera, indirect immunofluorescence Biochimica et biophysica acta Low 1477099
