Synthesis pass · prose summary of the discoveries below

MSP9 (PF3D7_1228600, also called ABRA/p101) is a P. falciparum merozoite surface and parasitophorous vacuole protein that contributes to erythrocyte invasion by directly engaging the host band 3 receptor (PMID:12076769, PMID:14630931, PMID:3042768). Its cysteine-rich N-terminal region binds the exofacial 5ABC region of erythrocyte band 3 specifically and saturably, selecting band 3 over glycophorin A/B and spectrin, with binding abolished by chymotrypsin pretreatment of erythrocytes; two regions of MSP9, Delta1 and Delta2, mediate this interaction and inhibit reinvasion in vitro (PMID:12076769, PMID:14630931). MSP9 does not act alone at the erythrocyte surface: it forms a stable co-ligand complex with MSP1(42), with the MSP9 Delta1a and Delta2 regions contacting the C-terminal MSP1(19)/MSP1(65) portions of MSP1, assembling a ternary MSP1–MSP9–band 3 assembly in which the same MSP9 regions used for complex assembly are those that bind band 3 (PMID:14630931, PMID:16289042, PMID:29511044). MSP9 is a broader component of the schizont/merozoite-stage MSP1-associated surface complex that also includes MSP3, MSP6, MSP7, and rhoptry proteins (PMID:21175202). Independently of its receptor-binding role, MSP9 possesses intrinsic chymotrypsin-like serine protease activity residing in its N-terminal portion and inhibited by chymostatin and PMSF but not leupeptin (PMID:10699251). Antibodies and synthetic peptides directed at the N-terminal region block merozoite reinvasion, marking MSP9 as an invasion-relevant, vaccine-candidate antigen (PMID:11483276, PMID:9596765).

2003 Two regions of MSP9 (MSP9/Delta1 and MSP9/Delta2) interact directly with the 5ABC exofacial region of erythrocyte band 3 during P. falciparum erythrocyte invasion. Soluble 5ABC blocked MSP9 binding to erythrocytes, and recombinant MSP9/Delta1 and MSP9/Delta2 inhibited P. falciparum reinvasion in vitro. Native MSP9 and MSP1(42) exist as a stable co-ligand complex that together targets the band 3 receptor. 2005 A major MSP9-binding site on MSP1 is located within the 19 kDa C-terminal domain (MSP1(19)). Two specific regions of MSP9, Delta1a and Delta2, interact with native MSP1(19) and with MSP1(42). The regions of MSP1 and MSP9 that bind the erythrocyte band 3 receptor are the same regions responsible for assembling the co-ligand complex, forming a ternary complex of MSP1–MSP9–band 3 during invasion. 2002 The amino terminus of MSP9 (also called ABRA) interacts with erythrocyte band 3 protein in a dose-dependent and saturable manner. Among band 3, glycophorin A, glycophorin B, and spectrin, only band 3 bound MSP9. The binding domain lies in the cysteine-rich N-proximal region of ABRA; binding was abolished by chymotrypsin treatment of erythrocytes. Native ABRA from P. falciparum-infected erythrocytes also bound band 3.
