SET10 (PF3D7_1221000) is a nuclear histone-lysine methyltransferase of Plasmodium falciparum whose principal in vivo catalytic activity is monomethylation of histone H3 at lysine 18 (H3K18me1); genetic deficiency abolishes H3K18me1 without reducing H3K4 methylation, revising the original assignment of SET10 as an H3K4-specific enzyme required for antigenic variation (PMID:39445823, PMID:22264509). SET10 localizes to the nucleus with peak abundance in schizonts and distinct subnuclear patterns that differ between asexual and sexual stages (PMID:19765590, PMID:39445823, PMID:39445826). It is dispensable for blood-stage viability, mutually exclusive var gene expression, var switching, sexual conversion, and gametocyte differentiation, but its loss reduces intraerythrocytic growth (PMID:33536326, PMID:39445823, PMID:39445826). Consistent with a repressive role for the H3K18me1 mark, SET10 knockout de-represses transcripts encoding red-blood-cell-remodeling and antigenic-variation proteins in schizonts (PMID:39445823). Proximity-labeling places SET10 within a broad nuclear chromatin-modulation network that includes ApiAP2 transcription factors, PfHDAC1, PfMORC, PfISWI, RNA polymerase II mediators, and DNA replication licensing factors (PMID:39445823).
