MDV1 (PF3D7_1216500), also called MDV-1/PEG3, is a sexual-stage-specific protein required for gametocyte maturation and gamete egress in Plasmodium (PMID:16275909, PMID:19438517). In P. falciparum it associates with the gametocyte plasma membrane, the parasitophorous vacuole membrane (PVM), and cleft-like membranous structures within the infected erythrocyte, and its disruption arrests most sexually committed parasites at stage I with disturbed membrane architecture and a pronounced loss of functional male gametocytes (PMID:16275909). MDV1 is also a resident protein of osmiophilic bodies, including male osmiophilic bodies (PMID:25262869, PMID:37492994); in P. berghei, knockout of the ortholog abolishes the ability of both male and female gametes to rupture the PVM and egress from the host erythrocyte, defining MDV1 as a mediator of a common, sex-shared egress mechanism distinct from asexual egress (PMID:19438517), with consequent failure of ookinete and oocyst development during mosquito transmission (PMID:19136003). Osmiophilic-body exocytosis marked by MDV1 precedes discharge of PPLP2-containing egress vesicles and, unlike the latter, is calcium-insensitive (PMID:37492994). MDV1 transcription is directly activated in P. falciparum by the ApiAP2 transcription factor PfAP2-G2, which binds the MDV1 transcriptional start site (PMID:33537025), and antibodies against MDV1 block parasite transmission to mosquitoes, marking it as a transmission-blocking antigen (PMID:34108248).

2005 MDV1 (PF3D7_1216500) protein in P. falciparum is sexual-stage specific and localizes to the gametocyte plasma membrane, parasitophorous vacuole membrane, and membranes of cleft-like structures within the erythrocyte. Disruption of pfmdv-1 causes dramatic reduction in mature gametocytes (especially functional male gametocytes), with most sexually committed parasites arrested at stage I, and produces disturbed membrane structures including multimembrane vesicles/tubes likely derived from deformed cleft-like structures. Gene disruption (knockout), immunofluorescence, electron microscopy, mosquito infectivity assay Proceedings of the National Academy of Sciences of the United States of America High 16275909
2007 MDV1 (PF3D7_1216500) protein in P. falciparum co-localizes with Pfs16 in the parasitophorous vacuole membrane and all derived membranous structures of early stage II gametocytes, including multi-laminate membrane whorls of circular clefts in the infected erythrocyte cytoplasm and membranes of gametocyte food vacuoles, as well as in the membrane and lumen of distinct cleft-like structures in the erythrocyte cytoplasm. Immunoelectron microscopy, immunofluorescence Experimental parasitology Medium 17367781
2009 MDV1 (PF3D7_1216500) ortholog in P. berghei (MDV-1/PEG3) is associated with osmiophilic bodies (dense granular organelles) in female gametocytes. Gene disruption of mdv-1/peg3 abolishes the ability of both male and female gametes to disrupt the parasitophorous vacuole membrane and egress from the host erythrocyte, resulting in strongly reduced zygote formation, establishing that MDV-1/PEG3 mediates a common egress mechanism shared by both sexes that is distinct from asexual parasite egress. 2008 MDV1 (PF3D7_1216500) ortholog in P. berghei is important for female gametocyte activation and ookinete development; knockout causes 86% reduction in ookinete conversion and 90% reduction in oocyst transmission. MDV-1 shows punctate cytoplasmic distribution in gametocytes, becomes peripherally located after activation, and in males concentrates into large foci. In developing ookinetes, MDV-1 localizes to the apical pole of the retort stage protrusion and to the posterior pole of fully developed ookinetes. 2014 MDV1 (PF3D7_1216500) ortholog in P. berghei localizes to both non-clustered and clustered male osmiophilic bodies (MOB), with clustering triggered by a drop in temperature independently of other stimuli; this localization pattern suggests that MDV1/PEG3 association with MOB precedes vesicle discharge during male gametocyte activation. 2021 The ApiAP2 transcription factor PfAP2-G2 (Pf3D7_1408200) directly regulates MDV1 (PF3D7_1216500) transcription in P. falciparum: PfAP2-G2 is enriched at the transcriptional start site of PfMDV-1 in ring and schizont stages (by ChIP-qPCR), and knockout of PfAP2-G2 leads to down-regulation of PfMDV-1 expression and a ~95% decrease in mature gametocytes with disproportionate loss of male gametocytes. 2023 MDV1 (PF3D7_1216500) is confirmed as a resident protein of osmiophilic bodies (OBs) in P. falciparum gametocytes; BioID proximity labeling and high-resolution imaging show that OB exocytosis (marked by MDV1/Peg3) precedes discharge of PPLP2-containing egress vesicles (P-EVs), and that OB exocytosis (unlike P-EV discharge) is calcium-insensitive. 2021 Antibodies raised against MDV1 (PF3D7_1216500) protein reduce oocyst counts in Anopheles mosquito midguts in a standard membrane feeding assay, demonstrating that MDV1 is a functional transmission-blocking antigen on the parasite surface accessible to antibody-mediated inhibition.
