PfORC1 (PF3D7_1203000) is a bifunctional nuclear protein of Plasmodium falciparum that couples DNA replication initiation to heterochromatic gene silencing through two functionally separable domains (PMID:16216221, PMID:18525026, PMID:22379140). Its C-terminal region carries an AAA+ nucleotide-binding fold with in vitro ATPase activity modeled on Cdc6/Cdc18 (PMID:16216221), and contains both a PCNA-interacting protein (PIP) motif that binds PCNA and an extreme C-terminal region that binds DNA directly; both C-terminal functions are conserved, as the DNA-binding region is interchangeable between P. falciparum and S. cerevisiae and the PIP motif is functional in yeast complementation (PMID:19633266). During asexual replication PfORC1 associates with putative autonomously replicating sequences (PfARSs) and colocalizes with PfPCNA and PfORC5 at trophozoite replication foci, marking replication origins (PMID:18554328, PMID:28644560). The unique N-terminal domain (aa 1–238) is independently targeted to the nuclear periphery and binds telomeric DNA via leucine heptad repeats, and full-length ORC1 occupies subtelomeric repeats and var gene promoters (PMID:22379140). This telomeric silencing function depends on Sir2: loss of PfSir2 severely compromises ORC1 spreading to subtelomeric regions, and the N-terminal domain functionally substitutes for yeast Sir3 in a Sir2-dependent manner (PMID:22379140, PMID:24018145). Cell-cycle progression is imposed on this activity by the CDK-like kinase PfPK5, which phosphorylates the N-terminal domain to abolish its DNA binding, alter subcellular localization, and trigger proteasome-mediated degradation of ORC1 at the schizont stage (PMID:26094711).

2005 PfORC1 (PF3D7_1203000) is expressed in the nucleus during late trophozoite and schizont stages, and its C-terminal region (aa 781-1033) displays ATPase activity in vitro, consistent with an AAA+ nucleotide-binding fold modeled on S. pombe Cdc6/Cdc18. 2008 PfORC1 (PF3D7_1203000) localizes to telomeric clusters at the nuclear periphery and the nucleolus. Prior to DNA replication, it dissociates from telomere clusters and spreads into the nucleus and cytoplasm. Chromatin immunoprecipitation showed physical association of ORC1 with telomere repeats but differential association with adjacent non-coding repeat DNA elements. ORC1 cooperates with Sir2 in telomeric silencing. Antibody localization (immunofluorescence), super gel-shift assay, chromatin immunoprecipitation (ChIP) Journal of cell science Medium 18525026
2008 PfORC1 (PF3D7_1203000) colocalizes with PfPCNA and PfORC5 at early-to-mid trophozoite replication foci, but is degraded at the late schizont stage, indicating stage-specific regulation of ORC1 during DNA replication. A putative PCNA-interacting protein (PIP) motif was identified in ORC1. Immunofluorescence colocalization, co-immunoprecipitation, western blot Molecular microbiology Medium 18554328
2009 The PIP domain in the C-terminus of PfORC1 (PF3D7_1203000) mediates physical interaction with PCNA in vitro and is functional in vivo (yeast complementation). The extreme C-terminal region directly binds DNA and can be swapped between S. cerevisiae and P. falciparum, indicating a conserved role in DNA replication. 2012 The unique N-terminus of PfORC1 (PF3D7_1203000; aa 1-238) is targeted to the nuclear periphery in vivo and binds telomeric DNA in vitro via leucine heptad repeats. Endogenous full-length ORC1 associates with subtelomeric repeat regions and var gene promoters. In PfSir2-deficient parasites, ORC1 binding and spreading to telomeric/subtelomeric regions is severely compromised, demonstrating ORC1 depends on Sir2 for localization (opposite to yeast where ORC is the Sir2 landing platform). Overexpression of ORC1-N(1-238) displaces Sir2 from subtelomeric loci and de-represses some var genes. 2013 PfORC1 (PF3D7_1203000) can functionally complement a Δsir3 mutant of S. cerevisiae for telomere silencing in a Sir2-dependent manner. The N-terminal domain (but not the first 70 aa) harbors the Sir3/ORC1-like silencing activity, as shown by chimera and deletion mutant analysis. 2015 PfORC1 (PF3D7_1203000) is phosphorylated at its N-terminal domain by the CDK-like kinase PfPK5. This phosphorylation abolishes DNA-binding activity of the N-terminus, leads to changes in subcellular localization, and triggers proteasome-mediated degradation of ORC1 at the schizont stage. These effects were confirmed using recombinant proteins in vitro and transgenic parasites expressing wild-type or phosphorylation-site mutant forms of ORC1. In vitro kinase assay with recombinant proteins, transgenic parasite expression of WT and mutant ORC1, subcellular localization imaging, proteasome inhibitor treatment, DNA-binding assay Molecular microbiology High 26094711

2022 PfORC1 (PF3D7_1203000) co-localizes with PfMYST at putative P. falciparum autonomously replicating sequences (PfARSs) during the trophozoite (replicating) stage, as demonstrated by ChIP, suggesting coordination between ORC1 and histone acetyltransferase activity at replication origins. Chromatin immunoprecipitation (ChIP) for ORC1 and PfMYST at PfARS loci, stage-specific analysis Experimental parasitology Low 36228701
2003 PfORC1 (PF3D7_1203000) is encoded by a single-copy gene on chromosome 12, contains all conserved ORC1/Cdc6/Cdc18 sequences, and its transcript (5600 nt) is expressed predominantly in the sexual erythrocytic stage (gametocytes), suggesting a role in DNA replication during gametogenesis.
