PF3D7_1107500 (PfPFD6) is a subunit of the Plasmodium falciparum prefoldin complex that functions in protein-substrate handling and has been implicated in artemisinin resistance (PMID:38925325). It has the capacity to assemble into a heteromeric prefoldin complex through systematic pairwise interactions with the other five PfPFD subunits (PFD1-6) (PMID:38925325). Together with all six prefoldin subunits, PfPFD6 associates with the artemisinin-resistance factor Kelch13, and expression of PfPFD6 in yeast PFD6-orthologous mutants reverts the phenotype to artemisinin resistance, establishing a causative genetic role for this subunit in the resistance mechanism (PMID:38925325). PfPFD6 engages client proteins, interacting with α-tubulin-I as a substrate, and its complex formation and interactions are blocked by the FDA-approved drug Biperiden, which suppresses growth of both artemisinin-sensitive (Pf3D7) and resistant (Pf3D7k13R539T) parasites (PMID:38925325). A separate report using pull-down and in silico analysis records a physical interaction with merozoite surface protein-1 (MSP-1) and expression at the trophozoite and schizont stages (PMID:33145997). Beyond these findings, no further mechanistic detail for PfPFD6 has been characterized in the available corpus.
