Mechanistic narrative

Synthesis pass · prose summary of the discoveries below

LSA1 (PF3D7_1036400) is a P. falciparum liver-stage-specific antigen expressed exclusively in infected hepatocytes during exo-erythrocytic development, where it functions in the maturation of the liver schizont (PMID:3306406, PMID:15784552). The ~200 kDa protein has a modular architecture: a central domain of tandem 17-amino-acid repeats flanked by conserved N- and C-terminal non-repetitive regions (PMID:1840628), and throughout liver schizogony the protein accumulates in the parasitophorous vacuole space (PMID:7515922). Its role is developmental rather than at parasite entry: targeted deletion of the locus in the NF54 strain leaves sporozoite gliding motility and hepatocyte invasion intact but causes a severe defect in late liver-stage schizogony and exo-erythrocytic merozoite formation, together with aberrant liver-stage expression of AMA-1 and circumsporozoite protein (PMID:21569184). The conserved N- and C-terminal regions and the repeat domain carry major B-cell and T-cell determinants, including an HLA-B53-restricted CD8+ CTL epitope recognized by malaria-immune donors, providing a molecular basis for the HLA-B53/severe malaria resistance association (PMID:1280333, PMID:7515922), and antibodies to LSA1 are a correlate of PfSPZ-CVac-induced sterile protection (PMID:41023197). Beyond these findings, the biochemical mechanism by which LSA1 controls liver-stage differentiation and the identity of its interacting partners have not been characterized in the available corpus.

1992 HLA-B53-restricted cytotoxic T lymphocytes (CTLs) from malaria-immune Africans recognized a conserved nonamer peptide from LSA1 (PF3D7_1036400), identifying an HLA-B53-restricted CTL epitope in this antigen and providing a molecular basis for the HLA-B53/severe malaria resistance association. 2003 Synthetic peptides from the LSA1 (PF3D7_1036400) sequence (peptides 20630, 20637, 20638, 20639) bind specifically and saturably to HepG2 hepatocyte cell membranes; peptide 20630 bound to a 48 kDa HepG2 surface membrane protein. 2011 Deletion of the lsa-1 locus in P. falciparum NF54 strain showed that LSA1 (PF3D7_1036400) is required for late liver-stage differentiation: lsa-1(-) sporozoites had normal gliding motility and hepatocyte invasion, but exhibited ~50% reduction in late liver-stage forms in a hepatocytic cell line at day 6, and a severe defect in late liver-stage schizogony and exo-erythrocytic merozoite formation in SCID/Alb-uPA mice with humanized livers at day 7. lsa-1(-) parasites also showed aberrant liver-stage expression of AMA-1 and circumsporozoite protein. 2005 LSA1 (PF3D7_1036400) is expressed solely in infected hepatocytes and is thought to have a role in liver schizogony and merozoite release; a recombinant construct (LSA-NRC) incorporating the N- and C-terminal regions plus two central 17-aa repeats, when expressed in E. coli and used to immunize rabbits and mice, elicited antibodies that recognized liver-stage parasites in infected human hepatocytes in culture, and induced IFN-γ-producing splenocytes in mice.
