PF3D7_1034700 encodes a class I ADP-ribosylation factor (PfARF1) that functions as a GDP/GTP-binding small GTPase regulating vesicular export from the endoplasmic reticulum in blood-stage Plasmodium falciparum (PMID:8954160, PMID:33604307). In vitro, recombinant PfARF1 is purified with bound GDP, binds GTP with first-order kinetics, lacks detectable intrinsic GTPase activity, and stimulates cholera-toxin-catalyzed ADP-ribosyltransferase activity, defining it as a class I ARF family member (PMID:8954160); its GDP-bound conformation was resolved crystallographically at 2.5 Å and resembles mammalian class I ARF1 (PMID:21045287). N-myristoylated PfARF1 localizes to the Golgi via its N-terminus, and its N-terminus alone is insufficient for parasite-plasma-membrane targeting or translocation into the parasitophorous vacuole (PMID:25909331). PfARF1 associates with PfRab5b and occupies ER subdomains distinct from PfRab1b, and both active and inactive PfARF1 mutants perturb ER exit, blocking trafficking of the PEXEL-negative PfAK2 to the parasitophorous vacuole membrane and reducing export of the PEXEL-positive protein Rifin (PMID:33604307). Biochemically, recombinant PfARF1 acts upstream of phosphoinositide signalling by activating the parasite's unique bifunctional PIP5K to promote PtdIns(4,5)P2 synthesis (PMID:19171150).
