Synthesis pass · prose summary of the discoveries below

PF3D7_1024600 (RAP-1) is a rhoptry protein of Plasmodium falciparum that functions as the organizing core of low-molecular-weight rhoptry complexes and is required for normal parasitophorous vacuolar membrane (PVM) structure and intra-erythrocytic growth after invasion (PMID:28205319). It is synthesized as an ~84–86 kDa precursor bearing a cleavable N-terminal signal peptide and is routed to the rhoptries through the classical secretory pathway, since brefeldin A, low temperature, and N-ethylmaleimide reversibly or irreversibly block processing of the precursor (PMID:8719244). Within the rhoptries the precursor undergoes two sequential N-terminal proteolytic cleavages to p82 and then p67, and these forms assemble into non-covalent complexes with RAP-2 and RAP-3 (PMID:9574915, PMID:17553576). Contrary to a role in invasion, conditional knockdown demonstrated that the RAP complex is dispensable for rhoptry morphology and erythrocyte invasion but is required post-invasion for normal PVM ultrastructure, growth kinetics, and in vivo parasitaemia (PMID:28205319); RAP-1 also serves as the determinant that tags non-parasitized erythrocytes with rhoptry proteins upon merozoite release (PMID:17624849). In P. berghei, RAP-1 is required to target RAP-2 to the rhoptries and contributes to virulence (PMID:29307564). Monoclonal antibodies against RAP-1 inhibit parasite growth and invasion in vitro, mapping to defined B-cell epitopes near the processing sites (PMID:2200961, PMID:1279419, PMID:9423886).

1990 RAP-1 (PF3D7_1024600) is a multi-component rhoptry antigen consisting of four major proteins (80, 65, 42, 40 kDa) and two minor proteins (77, 70 kDa); monoclonal antibodies against it inhibit parasite growth in vitro; the antigen is lipophilic and disulfide bonding plays an important role in its structure. Autocatalytic processing and immunological cross-reactivity suggest all components derive from the same precursor protein. Immunoaffinity purification, SDS-PAGE, Western blot, monoclonal antibody inhibition assay, gene sequencing Molecular and biochemical parasitology Medium 2200961
1992 Inhibitory monoclonal antibodies against RAP-1 map their epitopes to a region 10–20 amino acids C-terminal to the proteolytic cleavage site (at amino acid 190) responsible for processing of the 80-kDa form (p80) to the 65-kDa form (p65). The 65-kDa form (p65) is absent from purified merozoites, indicating its generation is associated with merozoite release rather than erythrocyte invasion. Beta-galactosidase fusion protein epitope mapping, synthetic peptide reactivity, Western blot, parasite growth inhibition assay Molecular and biochemical parasitology Medium 1279419
1995 Transport of RAP-1 (p82) to the rhoptries is regulated by the secretory pathway. Brefeldin A and low temperature reversibly block processing of the 86-kDa precursor (Pr86) to the mature 82-kDa product (p82). N-ethylmaleimide, which prevents transport vesicle fusion with Golgi membranes, irreversibly blocks Pr86 processing. In vitro translation indicates RAP-1 contains a cleavable N-terminal signal peptide and is initially synthesized as an ~84-kDa protein, suggesting an undetected protein modification occurs during maturation. Brefeldin A treatment, low-temperature block, N-ethylmaleimide treatment, in vitro translation, immunoprecipitation, metabolic labeling Molecular and biochemical parasitology High 8719244
1998 RAP-1 precursor Pr86 is processed to p82 and then to p67 by progressive removal of N-terminal epitopes in two sequential cleavage events within the rhoptries. Pr86 can form complexes containing RAP-2 product p39 and RAP-3 product p37, similar to complexes formed by mature p82/p67. Pr86 is present in rhoptries but not in segmenting schizonts or extracellular merozoites; p67 and p82 are the major forms in purified merozoites. Neither p67 nor p82 relocalize from rhoptries to the merozoite surface. Polyclonal and monoclonal antibody epitope mapping, immunoblot, size fractionation with brefeldin A, immunofluorescence, radiolabeling, purified merozoite analysis Molecular and biochemical parasitology High 9574915
1998 A second inhibitory B-cell epitope (iB-2) in RAP-1 was identified near the amino terminus of p67, distinct from the previously described iB-1 (N200TLTPLEELYPT211). Monoclonal antibody 1D6, which binds iB-2, inhibits parasite invasion of erythrocytes in vitro. Anti-iB-1 antibodies are immunogenic but not immunodominant and are not readily boosted upon reinfection in owl monkeys. 2007 RAP-1 is the core component of two low-molecular-weight rhoptry complexes (RAP1/RAP2 and RAP1/RAP3) identified in detergent-resistant membranes (DRMs) of P. falciparum schizonts. RAP-1 predominates in high molecular weight species by blue native PAGE, suggesting it aggregates in larger complexes than previously anticipated. 2007 RAP-1 and RAP-2 (but not RAP-3) bind to the surface of normal (non-parasitized) erythrocytes. Surface iodination experiments with RAP-1 gene-disruption mutant parasites demonstrated that RAP-1 is required for tagging of normal erythrocytes with rhoptry proteins after merozoite release. 2010 Pull-down experiments using anti-PfMSP-1 or anti-PfRhopH3 antibodies from merozoite lysate co-isolated PfRAP-1 and PfRAP-2 along with MSP-3, MSP-6, MSP-7, MSP-9, RhopH3, and RhopH1, identifying RAP-1 as part of a larger MSP-1-associated complex on the merozoite surface at schizont/merozoite stages. Antibody pull-down from merozoite lysate, tandem mass spectrometry, Western blot Journal of proteome research Low 21175202
2017 Conditional knockdown of RAP complex members (including RAP-1) in P. falciparum showed the RAP complex is not required for rhoptry morphology or erythrocyte invasion. Instead, post-invasion, the RAP complex is required for normal parasitophorous vacuolar membrane (PVM) structure and intra-erythrocytic parasite growth. RAP complex knockdown leads to altered PVM structure, delayed intra-erythrocytic growth, and reduced parasitaemias in infected mice. Conditional knockdown (DiCre or analogous system), electron microscopy of PVM, growth assays, mouse infection model Cellular microbiology High 28205319
2018 In P. berghei, RAP1 is required for targeting of RAP2 to the rhoptries. RAP1-deficient P. berghei ANKA parasites showed increased parasitemia during early infection but prolonged host survival. RAP1 contributes to virulence, as its deletion reduces lethality. 1992 Three matrix metalloproteinases (220, 95, and 70 kDa) co-isolate with the RAP-1 complex by non-covalent association, but these metalloproteinases originate from human serum used to cultivate P. falciparum in vitro rather than from the parasite itself.
