HSP60 (PfHsp60) was cloned from a P. falciparum blood-stage cDNA library; the deduced protein contains a putative mitochondrial targeting peptide at its amino-terminus and a GGM motif at its carboxyl-terminus. The hsp60 gene is located on chromosome 10. During heat shock, hsp60 transcript levels increase significantly in blood-stage parasites. |

PfHsp60 protein is localized to the mitochondrion of P. falciparum as determined by immunoelectron microscopy; immunofluorescence also detected PfHsp60 in the cytoplasm across all intraerythrocytic stages. Unlike PfHsp70, PfHsp60 protein expression was not induced by heat shock (37–40°C) despite a 3–4-fold increase in hsp60 mRNA, indicating a post-transcriptional block. Co-immunoprecipitation with anti-PfHsp60 antisera pulled down additional parasite proteins, suggesting interaction of PfHsp60 with other cellular proteins. |

The P. yoelii Hsp60 ortholog (PyHsp60) is encoded by a single-copy gene with a 201-bp intron 141 bp downstream of the ATG codon; its cDNA encodes a 579-amino-acid polypeptide with 93% amino acid identity to PfHsp60. A single, heat-inducible 2.3-kb transcript was detected in blood-stage parasites by Northern blot. Antisera raised against PyHsp60 recognized sporozoite, liver-stage, and blood-stage parasites, confirming expression across developmental stages. |

PfHsp60 was used as a mitochondrial marker in P. falciparum; immunofluorescence showed co-localization of PfIRPa with PfHsp60, establishing PfHsp60's utility as a bona fide mitochondrial marker protein in trophozoites. |

Precursor PfHsp60 accumulates in the parasite cytoplasm during ring, trophozoite, and schizont stages before being imported into the mitochondrion. Co-immunoprecipitation showed association of cytoplasmic precursor PfHsp60 with cytoplasmic chaperones PfHsp90 and PfHsp70-1. Metabolic pulse-chase labeling demonstrated translocation of the precursor pool into the mitochondrion over time. Geldanamycin (PfHsp90 inhibitor) treatment confirmed that precursor PfHsp60 is a client of PfHsp90 for its mitochondrial import. |

PfHsp60 potentially interacts with PfCalpain (a cysteine protease) in P. falciparum-infected erythrocytes. Co-immunoprecipitation and immunofluorescence assay indicated association of PfHsp60 and PfCalpain. Treatment with mizoribine (an Hsp60 inhibitor) attenuated PfCalpain enzyme activity and inhibited P. falciparum growth. |

In P. falciparum, hsp60 mRNA expression is co-ordinately regulated in a stage-dependent manner with antioxidant enzyme mRNAs: lowest at ring stage, highest at early trophozoite, and very low at schizont. Under oxidative stress (H2O2) or in G6PD-deficient RBCs, hsp60 mRNA expression is further up-regulated in a co-ordinated manner with antioxidant enzyme mRNAs, indicating redox-responsiveness of the hsp60 transcriptional program. |
