Mechanistic narrative

Synthesis pass · prose summary of the discoveries below

MRK (PF3D7_1014400) is a Plasmodium falciparum cyclin-dependent kinase most similar to human CDK7/MO15 that functions within the parasite nucleus to phosphorylate RNA polymerase II and DNA replication substrates (PMID:8944769, PMID:20332005). As a recombinant enzyme it displays intrinsic histone H1 kinase and autophosphorylation activity, forms a stable complex with human cyclin H that stimulates its activity, and is refractory to the classical CDK inhibitors olomoucine and roscovitine while being inhibited by human p21(CIP1) but not p16(INK4) (PMID:10717301, PMID:11700040). Its endogenous activator is PfMAT1, whose C-terminal domain physically associates with and stimulates MRK in a cyclin-dependent manner and confers substrate specificity to the kinase complex (PMID:16737745, PMID:20332005). Functionally, MRK phosphorylates the RNA polymerase II CTD but does not phosphorylate PfPK5, distinguishing it as a CTD kinase rather than a CDK-activating kinase, and phosphorylates the DNA replication proteins PfRFC-5 and PfMCM6 identified as interactors (PMID:16737745, PMID:20332005). Active-site mutations S138K and F143L render MRK constitutively active independently of cyclin or PfMAT1 (PMID:16737745). The kinase is transcribed predominantly in gametocytes yet was identified as likely essential for erythrocytic schizogony (PMID:8944769, PMID:22127061).

1996 MRK (PF3D7_1014400, referred to as Pfmrk in the abstract) is a single-copy gene on chromosome 10 encoding a 324-amino-acid CDK-like protein with highest similarity to human CDK7/MO15, containing two unique inserts (one of 5 amino acids before the cyclin-binding motif, one of 13 amino acids within the T-loop equivalent region); its 2500-nucleotide transcript is expressed predominantly in sexual blood stages (gametocytes). 2000 Recombinant MRK (PF3D7_1014400) expressed in E. coli exhibits histone H1 kinase activity and undergoes autophosphorylation in vitro; human cyclin H forms a stable complex with MRK and stimulates its kinase activity; olomoucine and roscovitine fail to inhibit MRK kinase activity. 2006 PfMAT1 (identified as an effector protein) stimulates MRK (PF3D7_1014400) kinase activity in a cyclin-dependent manner; PfMAT1 addition stimulates RNA polymerase II CTD phosphorylation by MRK but does not enable MRK to phosphorylate PfPK5, suggesting MRK functions as a CTD kinase rather than a CAK; active-site mutations S138K and F143L in MRK yield a 4–10-fold increase in kinase activity independently of cyclin or PfMAT1; MRK autophosphorylation is unaffected by cyclin or PfMAT1. In vitro kinase assays with recombinant proteins, active-site mutagenesis (S138K and F143L), CTD phosphorylation assay, substrate specificity assay Molecular and biochemical parasitology High 16737745
2010 MRK (PF3D7_1014400) physically interacts with PfMAT1 as shown by co-immunoprecipitation; the C-terminal domain of PfMAT1 is identified as the MRK activator domain; MRK and PfMAT1 co-localize to the parasite nucleus as demonstrated by immunofluorescence of GFP- and RFP-tagged versions; a bacterial two-hybrid screen identified DNA replication proteins PfRFC-5 and PfMCM6 as interactors with MRK; PfRFC-5 and PfMCM6 are substrates of MRK-mediated phosphorylation; PfMAT1 confers substrate specificity to the MRK kinase complex.
