Mechanistic narrative

Synthesis pass · prose summary of the discoveries below

ALBA3 (PF3D7_1006200) is a DNA/RNA-binding protein expressed across all intra-erythrocytic stages of Plasmodium falciparum that couples chromatin-level DNA binding to the control of subtelomeric virulence-gene expression (PMID:22006844, PMID:39868986). It binds DNA non-specifically at the minor groove and inhibits transcription in vitro, with this activity governed by reversible acetylation: acetylation of an N-terminal lysine lowers DNA-binding affinity, and the nuclear deacetylase PfSir2A interacts with ALBA3, removes this modification, and co-localizes with it at the nuclear periphery (PMID:22006844). ALBA3 occupies telomeric and subtelomeric chromatin, including var gene promoters, and also binds TARE6 subtelomeric repeats and single-stranded RNA, placing it at the interface of chromatin structure and RNA metabolism (PMID:22006844, PMID:22167473). Consistent with a regulatory role in virulence-gene control, episomal overexpression of ALBA3 impairs intra-erythrocytic growth and stage transitions and misregulates virulence-factor mRNAs while generally repressing var gene expression (PMID:39868986). In P. berghei gametocytes, ALBA3 is a component of the translationally silent DOZI/CITH mRNP complex, indicating an additional role in post-transcriptional mRNA storage (PMID:20169188). Beyond these findings, the structural basis of DNA binding and the direct molecular consequences of ALBA3 within the DOZI/CITH complex have not been characterized in the available corpus.

2011 ALBA3 (PfAlba3) binds to DNA non-specifically at the minor groove, and acetylation of its N-terminal lysine residue lowers its DNA-binding affinity. In vitro, PfAlba3 inhibits transcription by binding to DNA. PfSir2A (a nuclear-localized deacetylase) interacts with PfAlba3 and deacetylates its N-terminal lysine residue. PfAlba3 co-localizes with PfSir2A at the nuclear periphery and is found in telomeric and subtelomeric regions by FISH, ChIP, and ChIP-ReChIP analyses. It also binds to var gene promoters. The protein is expressed in all erythrocytic stages and exists predominantly in the acetylated form. DNA-binding assays (EMSA), in vitro transcription inhibition assay, acetylation/deacetylation assays, co-immunoprecipitation with PfSir2A, immunofluorescence/co-localization, FISH, ChIP, ChIP-ReChIP Nucleic acids research High 22006844
2011 ALBA3 (PfAlba3) is one of three PfAlba paralogs isolated as part of a molecular complex associated with the TARE6 subtelomeric region; EMSAs showed PfAlbas bind to TARE6 repeats and to single-stranded RNA, with different binding preferences among paralogs. In early blood stages PfAlba proteins are enriched at the nuclear periphery and partially co-localize with PfSir2; at the trophozoite–schizont transition they are also detectable in the cytoplasm in a punctate pattern. 2010 ALBA3 (PfAlba3) is identified as one of 16 major protein components of the DOZI/CITH messenger ribonucleoprotein (mRNP) complex purified from Plasmodium berghei gametocytes by affinity purification coupled with mass spectrometry. This mRNP contains translationally silent transcripts and integrates eIF4E and poly(A)-binding protein but excludes P-body RNA degradation factors and translation-initiation-promoting eIF4G. 2025 Ectopic overexpression of ALBA3 (PfAlba3) from an episome under a strong constitutive promoter strongly reduced P. falciparum intra-erythrocytic development (IED) growth and impaired stage transitions. RNA-seq of PfAlba3-overexpressing strains revealed significant misregulation of mRNAs encoding virulence factors (including erythrocyte invasion genes) and a general repression of var gene expression, linking ALBA3 to subtelomeric chromatin biology and mutually exclusive PfEMP1 expression.
