PF3D7_0917900 (BIP/PfGRP78) is an ER-resident HSP70-class molecular chaperone of Plasmodium falciparum, encoded by a gene expressed across asexual and sexual erythrocytic stages and homologous to mammalian GRP78 (PMID:3045815). Its ER residence is established directly by immuno-gold electron microscopy and by immunofluorescence, where it serves as a validated parasite ER marker distinct from the cis-Golgi, consistent with its non-canonical C-terminal SDEL retention signal (PMID:1779989, PMID:8223485). Unlike mammalian GRP78, BIP synthesis is not induced by heat shock, glucose deprivation, or classical ER-stress agents, defining a distinct stress-response profile (PMID:1779989). Functionally, BIP operates within the parasite ER quality-control machinery: it physically interacts with the ER co-chaperone PfGRP170 by reciprocal co-immunoprecipitation, and loss of PfGRP170 activates the eIF2α kinase PK4, placing BIP within the parasite unfolded protein response (PMID:31087747). BIP transcription is upregulated under artemisinin and stress conditions through direct binding of the histone acetyltransferase PfGCN5 at its locus, linking it to the artemisinin resistance response (PMID:33441725). Beyond the canonical ER pool, BIP partially localizes to the brefeldin A-sensitive Plasmodium export compartment and was identified as its 68-kDa component by mass spectrometry, implicating it in early steps of protein export to the host erythrocyte (PMID:12850257, PMID:32529065). The ATP-binding domain of BIP has been resolved crystallographically with ADP and with nucleoside-analogue fragment ligands, revealing parasite-specific active-site features exploitable for selective inhibition, and recombinant BIP is susceptible to chaperone inhibitors lethal to drug-sensitive and drug-resistant parasites, establishing it as a validated drug target (PMID:29985421, PMID:36275624).

PF3D7_0917900 (BIP/PfGRP78) was identified as a P. falciparum gene encoding a protein similar to the mammalian 78-kDa glucose-regulated protein (GRP78). The gene is expressed during asexual and sexual erythrocytic stages (RNA blot analysis), and a 72-kDa protein is immunoprecipitated from erythrocytic-stage parasites. The deduced protein sequence contains only three of the four canonical C-terminal ER-retention amino acids (Asp-Glu-Leu), suggesting possible ER localization. Genomic DNA cloning, RNA blot analysis, immunoprecipitation, sequence homology analysis Proceedings of the National Academy of Sciences of the United States of America Medium 3045815
PF3D7_0917900 (BIP/Pfgrp) localization was determined by immuno-gold electron microscopy to be primarily cytoplasmic, consistent with endoplasmic reticulum localization. Its synthesis was not induced by heat shock (unlike the cytosolic PfHsp70), nor by glucose deprivation, calcium ionophore A23187, tunicamycin, or 2-deoxyglucose, distinguishing its stress-response profile from canonical GRP78 inducers in mammalian cells. The C-terminal SDEL sequence was proposed to function similarly to KDEL in mammalian ER retention. Immuno-gold electron microscopy, metabolic labeling, Northern blot analysis, in vitro translation Molecular and biochemical parasitology Medium 1779989
PF3D7_0917900 (BIP) was used as a validated ER marker in P. falciparum. Upon treatment with brefeldin A (BFA), the localization of PfERD2 (normally in a single perinuclear Golgi focus) redistributed to a diffuse pattern similar to that of parasite BIP (ER marker), establishing that BIP resides in the parasite ER and that its localization is distinct from the cis-Golgi compartment. Indirect immunofluorescence, brefeldin A treatment, subcellular fractionation The EMBO journal Medium 8223485
PF3D7_0917900 (BIP/PfGRP78) expression was markedly induced in the hepatic (exoerythrocytic) stages of P. berghei (cultured in HepG2-A16 cells) and P. falciparum (in human hepatocytes in SCID mice), whereas expression was undetectable in sporozoites, establishing stage-specific upregulation of BIP during liver-stage development. 2003 PF3D7_0917900 (BIP) partially co-localizes with COPII coat proteins (Sar1p and Sec31p) and the 68 kDa and 22 kDa proteins of the Plasmodium export compartment (PEC), a brefeldin A-sensitive compartment proposed to be an early step in protein export from the parasite to the infected erythrocyte. This suggests the PEC is a domain of the ER or closely related to it. Co-localization immunofluorescence microscopy, brefeldin A treatment Molecular and biochemical parasitology Low 12850257
The crystal structure of the ATP-binding domain of P. falciparum GRP78 (PF3D7_0917900/BIP) was determined in complex with ADP. The structure revealed structural features unique to the parasite relative to human GRP78. Recombinant PfGRP78 was shown to be lethal to drug-sensitive and drug-resistant P. falciparum strains in vitro when inhibited by repurposed chaperone inhibitors (anticancer agents), and inhibitor binding to recombinant PfGRP78 was characterized biochemically, establishing BIP as a valid drug target. X-ray crystallography, recombinant protein binding characterization, in vitro parasite growth inhibition assay Scientific reports High 29985421
PF3D7_0917900 (BIP) physically interacts with the ER chaperone PfGRP170 in P. falciparum, as demonstrated by co-immunoprecipitation. Loss of PfGRP170 function (conditional mutant) led to activation of the P. falciparum eIF2α kinase PK4, placing BIP within the parasite unfolded protein response/ER stress pathway. BIP localizes to the parasite ER. Co-immunoprecipitation, conditional mutagenesis (TetR-DOZI system), immunofluorescence Cellular microbiology Medium 31087747
PF3D7_0917900 (BIP/PfHSP70-2) was identified as the 68 kDa protein recognized by a monoclonal antibody against the Plasmodium export compartment (PEC), by affinity purification and peptide mapping mass spectrometry. This localizes BIP to the PEC, a domain of the ER, and suggests a role for BIP in the export of Plasmodium proteins to the host erythrocyte. Affinity purification, peptide mapping by mass spectrometry, immunofluorescence Heliyon Medium 32529065
PfGCN5 (a histone acetyltransferase) was shown by ChIP-seq to directly bind to the BIP (PF3D7_0917900) gene locus in artemisinin-exposed P. falciparum, and BIP expression was upregulated under stress/artemisinin treatment conditions. This places BIP transcriptional regulation under epigenetic control via PfGCN5, linking BIP to the artemisinin resistance response. ChIP-seq (chromatin immunoprecipitation followed by sequencing), qRT-PCR expression analysis Scientific reports Medium 33441725
A high-throughput crystal-based fragment screening approach identified eight nucleoside analogue 'hit' compounds binding at the active site of the ATP-binding domain of P. falciparum GRP78 (PF3D7_0917900/BIP), as determined by X-ray crystallography. Binding was further validated by protein thermal shift assays. This established druggable sites on the ATP-binding domain of BIP distinct from those exploitable in human GRP78.
