Synthesis pass · prose summary of the discoveries below

CCp4 (PF3D7_0903800) is a multi-domain LCCL-domain adhesion protein of Plasmodium that functions within sexual-stage protein complexes required for parasite transmission through the mosquito (PMID:17950739, PMID:19304662). In P. falciparum, CCp4 is expressed on the surface of male and female gametocytes and macrogametes across gametocytogenesis stages II–V but is absent from exflagellating microgametes, and it co-localizes and forms a complex with the transmission-blocking antigen Pfs230 (PMID:17950739). It participates in larger PfCCp multi-protein complexes together with all other PfCCp family members, whose mutual stability is interdependent: loss of one member causes loss of the others at the protein but not mRNA level (PMID:19304662). These complexes are tethered to the gametocyte plasma membrane via Pfs230, which links to GPI-anchored Pfs48/45, and Pfs230 processing upon gametogenesis modulates complex association; disruption of Pfs230 causes proteolysis and release of the PfCCp proteins (PMID:26408859). Targeted disruption of CCp4 in P. falciparum does not block mosquito transmission (PMID:17950739). The P. berghei ortholog (PbLAP4) diverges functionally: it is translationally repressed in gametocytes, recruited post-fertilization to a pre-formed LAP1/2/3 complex through the SRCR modules of LAP1, localizes to the crystalloid organelle in ookinetes, and is essential for crystalloid biogenesis, oocyst cell-cycle regulation and sporozoite formation required for transmission (PMID:17335349, PMID:23684590, PMID:28414172, PMID:30367865).

2007 CCp4 (PfCCp4) is expressed on the surface of both male and female gametocytes and macrogametes (stages II–V of gametocytogenesis and early gamete formation) but is absent from exflagellating microgametes. CCp4 co-localizes with the transmission-blocking candidate Pfs230 and forms a protein complex with it. Targeted gene disruption of CCp4 does not inhibit parasite development in the mosquito vector, indicating a non-essential role during parasite transmission. Immunofluorescence assay (surface localization), co-immunoprecipitation (complex with Pfs230), targeted gene disruption with mosquito transmission phenotyping International journal for parasitology Medium 17950739
2009 CCp4 (PfCCp4) participates in multi-protein complexes (MPCs) with all other PfCCp family members in gametocyte lysates. Co-immunoprecipitation on gametocyte lysates demonstrated complex formation involving all six PfCCp proteins, and affinity chromatography co-elution binding assays with recombinant PfCCp domains indicated direct binding between distinct adhesion domains. Lack of one PfCCp protein leads to loss of other family members at the protein (but not mRNA) level. 2015 CCp4 (as part of PfCCp-based multi-protein complexes) is linked to the gametocyte plasma membrane via interactions with Pfs230, which itself binds GPI-anchored Pfs48/45. Upon gametogenesis (following mosquito uptake), GPI-anchored Pfs25 joins the MPC. Cleavage of Pfs230 at its N-terminal site upon gametogenesis increases its interaction with the MPC; loss of Pfs230 or impaired Pfs230 processing causes proteolysis of PfCCp proteins and their release from the MPC. 2022 Knockdown of the WD40-repeat protein PfWLP1 leads to reduced protein levels of PfCCp1 and PfCCp2 (key components of the PfCCp adhesion complex that includes CCp4), impaired gametocyte exflagellation, and reduced gametocyte numbers, indicating that PfWLP1 interaction with the PfCCp-based adhesion complex ensures complex stability. 2007 CCp4's P. berghei ortholog (PbLAP4) is critical for oocyst maturation and sporozoite formation; PbLAP4 null mutants fail to produce sporozoites and are not transmissible from mosquitoes to mice. The lethal sporozoite-formation phenotype is rescued only when the wild-type pblap4 allele is inherited from a female gametocyte, demonstrating that the relevant function is required before expression of the male-derived gene (i.e., in the gametocyte-to-ookinete period). Targeted gene disruption (loss-of-function mutant); genetic cross with wild-type, female-deficient (Δpbs47) and male-deficient (Δpbs48/45) parasites; mosquito transmission assay PLoS pathogens High 17335349
2013 CCp4's P. berghei ortholog (PbLAP4), like other LAP family members, localizes to the crystalloid organelle in ookinetes, as shown by GFP tagging. Unlike PbLAP1/2/3 which are expressed in gametocytes, PbLAP4 protein expression is not detected in gametocytes despite prominent transcription of the gene in sexual blood stages, indicating that translational repression controls its expression. 2017 CCp4's P. berghei ortholog (PbLAP4) is recruited to the LAP1/LAP2/LAP3 complex after fertilization. LAP1 facilitates recruitment of LAP4, LAP5, and LAP6 to the pre-formed LAP1/2/3 complex chiefly through LAP1's scavenger receptor cysteine-rich (SRCR) modules, as determined by affinity purification and quantitative mass spectrometry of reporter-tagged LAPs. 2018 A PbLAP4 mutant (corresponding to CCp4 ortholog in P. berghei) with abnormal crystalloid biogenesis produces oocysts with reduced growth and premature sporogony, providing evidence that the LAP complex (including PbLAP4) regulates oocyst cell division via the crystalloid. Targeted mutagenesis; microscopic phenotypic analysis of oocyst growth and sporozoite formation in transgenic P. berghei International journal for parasitology Medium 30367865
2020 CCp4's P. berghei ortholog (PbLAP4) is part of an extended LAP interactome of approximately fifty proteins identified by GFP-affinity pulldown of LAP3 combined with high-accuracy mass spectrometry; many of these interact proteins are also targeted to the crystalloid, indicating that the LAP complex (including PbLAP4) forms an intricate molecular assembly that facilitates crystalloid targeting and biogenesis.
