PF3D7_0804300 (MD3) is a putative CCCH-type zinc finger protein that acts as a post-transcriptional regulator driving male gametocyte development in Plasmodium falciparum (PMID:38148574, PMID:42244567). It localizes to the cytoplasm of immature male gametocytes, where its disruption impairs gametocyte maturation and male gametocytogenesis (PMID:38148574). MD3 binds male gamete-associated mRNA transcripts through a specific RNA sequence motif and promotes their translation, with proteins encoded by MD3-bound transcripts significantly depleted in knockout parasites; loss of MD3 causes a male fertility defect and reduced mosquito infectivity (PMID:42244567). Consistent with a translational regulatory role, MD3 assembles into a post-transcriptional regulatory complex, interacting with the RNA-binding proteins PABP1 and ALBA3, translation initiation and repression factors eIF4G, PUF1, NOT1 and CITH, and further gametocytogenesis regulators ZNF4, MD1 and GD1 (PMID:38148574). In the rodent model P. berghei, the MD3 ortholog is essential for male sexual development and functions within the regulatory cascade governing male gametocyte determination and differentiation (PMID:36634679). Beyond these findings, no additional mechanistic detail for PF3D7_0804300 has been characterized in the available corpus.
