PF3D7_0604800 (RAP-1) is a rhoptry protein that serves as the organizing core of low-molecular-weight rhoptry complexes required for post-invasion intra-erythrocytic development in Plasmodium falciparum (PMID:17553576, PMID:28205319). It is synthesized as an 86-kDa precursor (Pr86) bearing a cleavable N-terminal signal peptide and is transported to the rhoptries through the secretory pathway, as its precursor-to-mature processing is reversibly blocked by brefeldin A and low temperature and irreversibly blocked by N-ethylmaleimide (PMID:8719244). Within the rhoptries Pr86 undergoes sequential proteolytic maturation, first rapidly to p82 and then more slowly to p67, with these mature forms — and not the surface — being the species retained in released merozoites (PMID:2200961, PMID:9574915). RAP-1 nucleates two distinct complexes with RAP-2 and RAP-3 respectively, which partition into detergent-resistant membranes (PMID:9574915, PMID:17553576), and in P. berghei RAP-1 is required to target RAP-2 to the rhoptries (PMID:29307564). RAP-1 can bind the surface of normal erythrocytes (PMID:17624849), yet the RAP complex is dispensable for rhoptry morphology and erythrocyte invasion; instead, conditional knockdown shows it is required after invasion for normal parasitophorous vacuolar membrane structure, efficient intra-erythrocytic growth, and virulence in mice (PMID:28205319, PMID:29307564).

1990 RAP-1 is a lipophilic antigen in which disulfide bonding plays an important role in its structure. The multi-component antigen (80, 65, 42, 40 kDa plus minor components of 77, 70 kDa) is derived from a single precursor protein, as suggested by monoclonal antibody evidence, autocatalytic processing, and immunological cross-reactivity among all components. Immunoaffinity purification, SDS-PAGE, Western blot, autocatalytic processing assays, monoclonal antibody epitope analysis Molecular and biochemical parasitology Medium 2200961
1992 Inhibitory monoclonal antibodies against RAP-1 map their epitopes to a region 10–20 amino acids C-terminal to the proteolytic cleavage site (at amino acid 190) where p80 is processed to p65. The p65 form is absent from purified merozoites, indicating its generation is associated with merozoite release rather than erythrocyte invasion. 1995 Transport of RAP-1 to the rhoptries is regulated by the secretory pathway. Brefeldin A (BFA) and low temperature reversibly block processing of the 86-kDa precursor (Pr86) to the mature 82-kDa RAP-1 product (p82). N-ethylmaleimide, which prevents vesicle-Golgi fusion, irreversibly blocks Pr86 processing. In vitro translation revealed a cleavable N-terminal signal peptide and an initial 84-kDa primary translation product, distinct from the in vivo Pr86 precursor, indicating an additional protein modification during maturation. Brefeldin A treatment, low-temperature block, N-ethylmaleimide treatment, in vitro translation, metabolic labeling, immunoprecipitation Molecular and biochemical parasitology High 8719244
1998 RAP-1 precursor Pr86 is processed in the rhoptries by two sequential cleavage events: rapid cleavage to p82 followed by delayed cleavage to p67. Pr86, RAP-2 product p39, and RAP-3 product p37 form complexes prior to processing. Pr86 is present transiently in rhoptries and is absent from segmenting schizonts and extracellular merozoites, whereas p67 and p82 are the major RAP-1 forms in purified merozoites. Neither p67 nor p82 relocalize from rhoptries to the merozoite surface. Immunoblot with epitope-mapped antibodies, size fractionation with BFA-blocked radiolabeling, immunofluorescence localization, purified merozoite analysis Molecular and biochemical parasitology High 9574915
2007 RAP-1 is the core component of two distinct low-molecular-weight rhoptry complexes (RAP1/RAP2 and RAP1/RAP3) that partition into detergent-resistant membranes (DRMs) of P. falciparum schizonts. RAP-1 predominates in high-molecular-weight species by blue native PAGE, suggesting aggregation into larger complexes than previously anticipated. Detergent-resistant membrane purification, blue native PAGE, mass spectrometry identification Molecular and biochemical parasitology Medium 17553576
2007 RAP-1 and RAP-2 (but not RAP-3) bind to the surface of normal (non-parasitized) erythrocytes. This was demonstrated using RAP-1 gene-disruption mutant parasites in which the capacity to tag erythrocytes with surface iodination was lost, directly implicating RAP-1 in erythrocyte surface binding. 2017 The RAP complex (including RAP-1) is not required for rhoptry morphology or erythrocyte invasion, but is required post-invasion for normal parasitophorous vacuolar membrane (PVM) structure and intra-erythrocytic parasite growth. Conditional knockdown of RAP complex members leads to altered PVM structure, delayed intra-erythrocytic growth, and reduced parasitemia in infected mice. Conditional knockdown (reverse genetics), electron microscopy of PVM, parasite growth assays, mouse infection model Cellular microbiology High 28205319
2018 In P. berghei, RAP-1 is required for targeting RAP-2 to the rhoptries. RAP1-deficient parasites showed increased early parasitemia but prolonged host survival, while RAP2/3-deficient parasites showed low parasitemia and host recovery, indicating RAP-1 and RAP-2/3 contribute to virulence. Generation of RAP1-deficient and RAP2/3-deficient P. berghei mutants by reverse genetics, mouse infection experiments, complementation of RAP2/3 expression Experimental parasitology Medium 29307564
2010 RAP-1 was identified as part of an MSP-1-associated complex on the merozoite surface by anti-MSP-1 and anti-RhopH3 antibody pulldowns from merozoite lysates, with the complex detected at schizont/merozoite stages but not ring stages.
