Synthesis pass · prose summary of the discoveries below

PF3D7_0501100 (PFE0055c) is an exported P. falciparum type II J-domain co-chaperone that is trafficked into the cytosol of the infected human erythrocyte, where it localizes to novel punctate, highly mobile, cholesterol-sensitive membrane-associated structures termed J-dots that are distinct from Maurer's clefts (PMID:20482550). Its export to the host cell cytosol was independently confirmed by a self-assembling split-GFP compartmentalization assay (PMID:23271009), and its predicted substrate-binding domain is both necessary and sufficient for J-dot targeting; this targeting operates in P. falciparum-infected but not P. berghei-infected erythrocytes, indicating a P. falciparum-specific mechanism (PMID:27063072). Functionally, recombinant PFE0055c acts as a co-chaperone by stimulating the basal ATPase activity of the exported malarial chaperone PfHsp70-x through its J domain, an interaction blocked by the J-domain inhibitor C86, while its isolated J domain promiscuously stimulates PfHsp70-x as well as human HSP70 and HSC70 in vitro (PMID:33236291, PMID:41618743). Within J-dots it associates with PfHsp70-x and the co-chaperone PFA0660w (PMID:34569022). Beyond this exported co-chaperone role and its partnership with PfHsp70-x, no direct client substrate of PF3D7_0501100 has been characterized in the available corpus.

  1. 2010 Medium

Establishing where an exported J-domain protein resides in the host cell was the first step in defining its role; PFE0055c was found in a previously unrecognized mobile, cholesterol-dependent compartment, defining the J-dot as a distinct trafficking structure.

  3. 2016 Medium

To understand how PFE0055c reaches J-dots, domain dissection identified the substrate-binding domain as necessary and sufficient for targeting, and cross-species comparison established that this targeting is P. falciparum-specific.

2010 PFE0055c (PF3D7_0501100), when expressed as a GFP fusion in P. falciparum-infected erythrocytes, localizes to novel punctate mobile structures distinct from Maurer's clefts, termed 'J-dots'. These structures are tightly membrane-associated and sensitive to cholesterol depletion, suggesting interaction with a cholesterol-rich membrane binding partner, and are proposed to be involved in trafficking parasite-encoded proteins through the erythrocyte cytosol. 2012 Using a self-assembling split-GFP (saGFP) system in transfected P. falciparum-infected erythrocytes, PFE55 (PF3D7_0501100) was confirmed to be exported to the host erythrocyte cytosol (compartmentalization verified without reliance solely on fluorescent phenotype). 2016 The predicted substrate-binding domain of PFE0055c (PF3D7_0501100) is necessary and sufficient for targeting the protein to J-dots in P. falciparum-infected human erythrocytes. This J-dot targeting is not conserved when the protein is expressed in P. berghei-infected murine erythrocytes, indicating J-dots and their targeting mechanism are P. falciparum-specific. 2020 Recombinant full-length PFE0055c (PF3D7_0501100) stimulates the basal ATPase activity of the exported chaperone PfHsp70-x to a greater extent than the co-chaperone PFA0660w, but does not significantly stimulate the ATPase activity of human Hsp70. The J-domain inhibitor C86 (chalcone) specifically inhibits the PFE0055c-stimulated ATPase activity of PfHsp70-x (but not its basal activity), consistent with PFE0055c binding to PfHsp70-x through its J domain. Benzothiazole rhodacyanines JG231 and JG98 inhibit both basal and PFE0055c-stimulated ATPase activity of PfHsp70-x. 2023 In silico virtual screening identified small molecules (zoliflodacin, itraconazole, ezetimibe, benzo-diazepinone) predicted to disrupt or modulate the PfHsp70-x–PFE0055c (PF3D7_0501100) chaperone complex, with preferential binding affinity for the malarial system over the human system validated by molecular dynamics simulation. Two classes of modulators were distinguished: those inhibiting complex formation and those stabilizing it. 2026 The isolated J-domain of PFE0055c (PF3D7_0501100) stimulates the ATPase activity of PfHSP70-X, human HsHSP70, and human HsHSC70 in vitro, demonstrating promiscuous stimulation across HSP70 family members, though differences in stimulation magnitude were observed between individual PfJDP–HSP70 pairings.
