Synthesis pass · prose summary of the discoveries below

Pfs16 (PF3D7_0406200) is a 16-kDa, 157-amino-acid integral membrane protein of the parasitophorous vacuole membrane (PVM) that serves as one of the earliest markers of sexual commitment in Plasmodium falciparum, with expression induced immediately after invasion by sexually committed merozoites and sustained through gametocytogenesis and into macrogametes and sporozoites (PMID:7935618, PMID:2038355, PMID:9364968). Its expression is switched on transcriptionally at the onset of gametocytogenesis via a defined promoter activated in sexually committed ring stages, with additional post-transcriptional modulation (PMID:9364968, PMID:9891033). PVM targeting and retention are encoded by its C-terminal 53 amino acids, which carry a motif shared with other PVM proteins, while the region between the signal sequence and transmembrane domain drives 'capping'—concentration of the protein at the gametocyte ends during emergence from the PVM and red blood cell (PMID:21498641); structurally it is an alpha-helical protein spanning the PVM through a single transmembrane domain (PMID:37196765). Targeted disruption of Pfs16 is not lethal to sexual development but reduces gametocyte production, impairs male exflagellation, and produces a truncated mislocalized protein, establishing a role in optimal gametocyte formation (PMID:14698439). Beyond the parasite, recombinant Pfs16 binds Anopheles gambiae midgut epithelial cells and suppresses mosquito caspase 3/7 activity to facilitate transmission, and anti-Pfs16 antibodies reduce oocyst numbers and block transmission (PMID:37196765, PMID:34262880). Pfs16 is the direct molecular target of the transmission-blocking N-4HCS sulphonamide compound class, whose treatment phenocopies genetic knockout (PMID:36715290).

1994 Pfs16 is localized to the parasitophorous vacuole membrane (PVM) of P. falciparum gametocytes. Purification of PVMs from mature and immature gametocytes combined with immunoelectron microscopy established this localization. During gametogenesis, the PVM is shed along with the RBC membrane, and Pfs16 is retained on the disintegrating membrane; in a minority of cases, Pfs16 can be retained on the gamete surface. First expression was estimated at 35 hours post-merozoite invasion in sexually committed ring stages. Immunofluorescence assay, immunoelectron microscopy, PVM purification from gametocytes Molecular and biochemical parasitology High 7935618
1994 Pfs16 is localized specifically to the parasitophorous vacuole membrane (PVM) in gametocytes, and during gametogenesis when the PVM disintegrates, the antigen is detected on multilaminated whorls (remains of the PVM) and not on the gamete plasma membrane. Pfs16 is also present on membranes surrounding cytosomes and food vacuoles within the parasite cytoplasm, and on structures believed to be Garnham bodies in the host cell cytoplasm. Immunoelectron microscopy Parasitology High 8159458
1991 Pfs16 encodes a 157-amino-acid, 16-kDa integral membrane protein with a putative N-terminal signal sequence, a hydrophobic membrane anchor, and a highly hydrophilic C-terminal region. The protein is expressed on the surface of macrogametes and sporozoites but not in asexual erythrocytic stages, as demonstrated by immunofluorescence and immunoelectron microscopy with protein A-gold. cDNA cloning, Western blot, immunofluorescence assay, immunoelectron microscopy (protein A-gold) Molecular and biochemical parasitology High 2038355
1997 Expression of Pfs16 is induced immediately following invasion of a red blood cell by sexually committed merozoites and continues throughout gametocytogenesis and in macrogametes. Regulation is at the level of transcription initiation and is also modulated by a post-transcriptional process, as shown by comparative analysis of promoter activity, RNA levels (in situ hybridization at single-cell level), and de novo protein synthesis rates. Promoter-reporter transfection assay, Northern blot, in situ hybridization, metabolic labeling of de novo protein synthesis Molecular and biochemical parasitology High 9364968
1999 The pfs16 promoter is activated at the onset of gametocytogenesis (in sexually committed ring-stage parasites), marking the earliest developmental switch in sexual differentiation. The regions of the pfs16 promoter essential for high transcriptional activity were identified by deletion analysis using transfection of P. falciparum and P. gallinaceum. Promoter deletion analysis, P. falciparum and P. gallinaceum transfection with reporter constructs Molecular and cellular biology High 9891033
2004 Targeted disruption of the Pfs16 gene by homologous recombination (three independent clones confirmed) resulted in: (1) absence of full-length Pfs16 transcripts and PVM-localized 16 kDa protein; (2) expression of a 14 kDa truncated protein mislocalized to an unidentified double-membrane compartment; (3) significant reduction in gametocyte production; (4) preliminary evidence of impaired male gametocyte exflagellation in vitro. Pfs16 is not essential for sexual development but is required for optimal gametocyte production. 2007 Pfs16 and Pfpeg3/mdv1 proteins co-localize in the PVM and in all derived membranous structures of stage II gametocytes, including multi-laminate membrane whorls of circular clefts in the erythrocyte cytoplasm, membranes of food vacuoles, and distinct cleft-like structures in the erythrocyte cytoplasm, as shown by immunoelectron microscopy. 2011 The 53 C-terminal amino acids of Pfs16 (appended to a GFP reporter already containing the N-terminal secretory signal) are sufficient for targeting to and retention on the PVM. Removal of the 11 C-terminal amino acids did not prevent PVM targeting but reduced membrane retention. A sequence motif identified in the C-terminal targeting region is shared with seven other known PVM proteins. Additionally, the region between the N-terminal signal sequence and the transmembrane domain (not required for PVM targeting) was found to be involved in 'capping'—concentration of native Pfs16 and full-length Pfs16-GFP at the ends of the gametocyte during emergence from the PVM and RBC. 2018 The P. berghei syntenic gene PBANKA_1003900 (which has a similar domain structure to Pfs16 but low overall sequence similarity) is expressed only in gametocytes and ookinetes. A knockout mutant of PBANKA_1003900 in P. berghei showed no defect in sexual development or ookinete formation, though a reduction in oocyst number was observed in mosquitoes. 2023 Photoaffinity labeling of a photoactivatable probe derivative of the N-[(4-hydroxychroman-4-yl)methyl]-sulphonamide compound DDD01035881 coupled with mass spectrometry identified Pfs16 as a direct binding target. Cellular thermal shift assay confirmed that the parent compound DDD01035881 stabilizes Pfs16 in lysates from activated mature gametocytes. Parasites treated with N-4HCS compounds phenocopied the targeted deletion of Pfs16 in gametocytes (by high-resolution fluorescence and electron microscopy), implicating Pfs16 as the functional target of this transmission-blocking compound class. 2023 Recombinant Pfs16 (insect-cell expressed) interacts directly with Anopheles gambiae midgut lysate as shown by ELISA, and microscopy demonstrated that recombinant Pfs16 binds to midgut epithelial cells. Feeding recombinant Pfs16 to mosquitoes increased oocyst numbers. Pfs16 reduced caspase 3/7 activity in mosquito midguts, implicating it in suppression of the mosquito Jun-N-terminal kinase immune pathway.
