P230 (PF3D7_0209000) is a large 6-cysteine family surface protein that acts as a gamete surface factor required for fertilisation and mosquito transmission of Plasmodium (PMID:16879650, PMID:20386715). In gametocytes it is expressed as a ~360 kDa protein localised to the parasitophorous vacuole/parasite plasma membrane, and upon gamete emergence it is proteolytically cleaved at its amino-terminus to a 310 kDa surface-exposed form, shedding a 50 kDa fragment that contains an immunodominant glutamate-rich repeat region (PMID:8813686). On the gamete surface P230 assembles with the 48/45 kDa glycoprotein into a complex stabilised in part by disulfide bonds and sensitive to chaotropic agents, pH extremes, and anti-P230 antibodies (PMID:1380124), yet P230 (which has no known membrane anchor) can persist on the membrane surface independently of Pfs48/45 (PMID:32709983). Functionally, P230 mediates red-blood-cell binding by exflagellating male gametes: gene-disruption mutants exflagellate normally but fail to form exflagellation centres, and show sharply reduced oocyst production and mosquito infectivity (PMID:16879650); in P. berghei the ortholog is a male fertility factor mediating gamete recognition and attachment (PMID:20386715) and, together with P48/45, enforces pre-zygotic reproductive isolation between Plasmodium species (PMID:25854886). The N-terminal cysteine-motif domain 1 adopts a 6-cysteine/SAG1-like fold and harbours the principal transmission-blocking epitope: antibodies against domain 1 confer transmission-blocking activity in membrane-feeding assays (PMID:30824357), and a complement-dependent human monoclonal antibody binds a large, conserved discontinuous conformational epitope within this domain (PMID:32709983, PMID:33741942).

2006 P230 (Pfs230) mediates RBC binding by exflagellating male gametes: Pfs230 gene-disruption mutants successfully exflagellate but are unable to interact with red blood cells and form exflagellation centres, demonstrating that P230 is the surface molecule on male gametes responsible for RBC binding during exflagellation. Gene disruption (two independent clones with distinct Pfs230 gene disruptions) followed by exflagellation and mosquito infectivity assays Molecular microbiology High 16879650
2006 P230 (Pfs230) is required for oocyst development and mosquito infectivity: Pfs230 disruptants showed 92–96% reduction in oocyst production and 71–76% reduction in mosquito infectivity, independent of Pfs48/45 co-expression. Gene disruption with standard membrane feeding assay (SMFA) and mosquito infectivity assay Molecular microbiology High 16879650
2010 P230 is a male fertility factor in Plasmodium berghei: genetic disruption of p230 abolishes gamete recognition and attachment, reducing fertilisation, in a manner analogous to disruption of P48/45, and consistent with P230 and P48/45 forming a protein complex. 1996 P230 (Pfs230) undergoes stage-specific proteolytic processing: it is expressed as a 360 kDa protein localised to the parasitophorous vacuole/parasite plasma membrane in gametocytes, and upon gamete emergence it is cleaved at the amino-terminus to a 310 kDa surface-exposed form, with the removed 50 kDa fragment containing an immunodominant repeat region (25 contiguous glutamates and an EEVG16 repeat). 1992 P230 (the 230 kDa gamete surface protein) forms a protein complex with the 48/45 kDa glycoproteins on the gamete surface; this complex is disrupted by chaotropic agents, extremes of pH, and by antibodies against the 230 kDa protein (but not by antibodies against the 48/45 kDa doublet). Disulfide bonds contribute to complex integrity, as chemical modification of disulfide/sulfhydryl groups (requiring prior denaturation) perturbs the interaction. 2020 Crystal structure of recombinant Pfs230 domain 1 (Pfs230D1M) was determined in complex with Fab fragment of transmission-blocking mAb 4F12, revealing a 6-cysteine domain structure. The arrangement of P230 on the surface of macrogametes differs from that on microgametes, and P230 (with no known membrane anchor) can exist on the membrane surface in the absence of Pfs48/45. 2021 A transmission-blocking human monoclonal antibody against P230 domain 1 acts in a complement-dependent manner and binds a large discontinuous conformational epitope comprising six structural elements within domain 1 of P230; this epitope is conserved across field isolates. 2015 P230 is a key determinant of pre-zygotic reproductive isolation between Plasmodium species: female gametes lacking P230 (or P48/45) show dramatically elevated hybridisation rates with heterospecific males, and positive selection acts on a region of P230, consistent with its role as a gamete recognition protein. Genetic knockout of p230 (and p48/45) in P. berghei and P. yoelii, mixed-species mating assays, molecular evolution analysis Proceedings. Biological sciences Medium 25854886
2005 Structural modelling of Pfs230 double domains reveals fold resemblance to SAG1 (a double beta-sandwich surface protein from Toxoplasma gondii), and template-directed modelling onto SAG1 places cysteine positions consistent with predicted disulfide-bonding arrangements; a highly clustered polymorphism region within the second double domain maps to one face of the sandwich, suggesting a functional surface. Fold prediction and template-directed homology modelling onto SAG1 structure Proceedings of the National Academy of Sciences of the United States of America Low 16155126
2019 Antibodies conferring transmission-blocking activity against P230 (Pfs230) map exclusively to cysteine motif (CM) domain 1: all recombinant Pfs230 constructs containing CM domain 1 induced strong inhibition in standard membrane feeding assays, whereas constructs lacking CM domain 1 showed no inhibition, establishing domain 1 as the primary functional epitope region.
