SERA6 (PF3D7_0207500) is an essential parasitophorous-vacuole cysteine protease of blood-stage Plasmodium falciparum that executes the terminal step of merozoite egress by dismantling the host red blood cell membrane cytoskeleton (PMID:29459732). First identified as a schizont-stage 130-kDa polypeptide localized within the parasitophorous vacuole and bearing a cysteine-proteinase-homologous domain (PMID:1840623), SERA6 is refractory to genetic disruption and to substitution of its predicted catalytic cysteine, establishing that both the gene and its enzymatic activity are essential for the erythrocytic cycle (PMID:12228245, PMID:22984267). SERA6 is activated through an ordered two-step proteolytic cascade: the parasitophorous-vacuole serine protease SUB1, discharged from exonemes just prior to egress, cleaves SERA6 (PMID:18083098, PMID:22984267), triggering a second autocatalytic maturation step that requires assembly with the SUB1-substrate cofactor MSA180 in multimolecular complexes (PMID:33932049). Mature SERA6 then cleaves β-spectrin within its actin-binding domain to disassemble the RBC membrane; SERA6-null parasites rupture the parasitophorous vacuole membrane normally but fail to rupture the erythrocyte membrane, defining SERA6's role as specific for RBCM disassembly (PMID:29459732). Cell-free reconstitution confirms strict MSA180-dependence of SUB1-triggered autoprocessing and demonstrates that fully mature SERA6 cleaves a synthetic peptide based on its autocatalytic site, and small-molecule inhibitors of SERA6 autoprocessing block egress (PMID:33932049, PMID:42662414). The pathway is conserved across species: the P. berghei ortholog is essential for hepatic merozoite egress and, unlike the vacuolar blood-stage enzyme, localizes to the ookinete and sporozoite surface (PMID:41616267).

2007 PfSUB1, discharged from exonemes into the parasitophorous vacuole just prior to egress, mediates proteolytic maturation of SERA6 (along with at least one other SERA family member); pharmacological blockade of PfSUB1 inhibits egress and ablates invasive capacity of merozoites. Subcellular fractionation, immunofluorescence, protease inhibitor pharmacology, Western blot of cleavage products Cell High 18083098
2002 SERA6 is expressed as a protein in blood-stage parasites and is essential for the erythrocytic cycle; repeated attempts to disrupt the sera6 locus by homologous recombination failed, indicating the gene is refractory to knockout. Reverse transcriptase-PCR, microarray, Western blot with SERA6-specific antibodies, attempted gene disruption by homologous recombination The Journal of biological chemistry High 12228245
2012 SERA6 localizes to the parasitophorous vacuole (PV) and is precisely cleaved just prior to egress by the serine protease PfSUB1; this cleavage activates SERA6 as a cysteine protease. Mutations replacing the predicted catalytic Cys of SERA6, or blocking PfSUB1 processing, could not be stably introduced into the parasite, confirming that enzymatic activity and processing are both essential. Immunofluorescence localization, Western blot of processing products, active-site mutagenesis (attempted stable integration), in vitro cysteine protease activity assay after SUB1 cleavage The Journal of biological chemistry High 22984267
2018 SERA6 is required specifically for rupture of the red blood cell membrane (RBCM) but not for PVM rupture or RBCM poration; SERA6-null parasites rupture the PVM normally but fail to rupture the RBCM. SERA6 requires SUB1-mediated processing to function. SERA6-dependent proteolytic cleavage within the actin-binding domain of β-spectrin mediates RBCM disassembly. Conditional SERA6 gene deletion (complementation studies), live-cell and cryo-electron microscopy of egress morphology, Western blot of β-spectrin cleavage products in SERA6-null vs. wild-type parasites Nature microbiology High 29459732
2021 SERA6 activation involves a second autocatalytic step triggered by SUB1 cleavage; this autoproteolytic maturation requires interaction in multimolecular complexes with a distinct PV-located protein cofactor called MSA180, which is itself a SUB1 substrate. Genetic ablation of MSA180 phenocopies SERA6 disruption (fatal block in β-spectrin cleavage and RBC rupture). Drug-like inhibitors of SERA6 autoprocessing block β-spectrin cleavage and egress in both P. falciparum and P. knowlesi. Co-immunoprecipitation/pull-down of SERA6–MSA180 complexes, conditional genetic ablation of MSA180, Western blot of SERA6 autocleavage and β-spectrin cleavage, small-molecule inhibitor assays (egress block), cross-species validation in P. knowlesi The EMBO journal High 33932049
1991 SERA6 (described as SERPH / 130-kDa protein) is a schizont-stage polypeptide localized within the parasitophorous vacuole, detected by Western blot and immunoelectron microscopy; its domain homologous to cysteine proteinases suggested a possible protease role at merozoite release. 2026 A cell-free in vitro system recapitulates SERA6 maturation, confirming strict requirement for MSA180 in SUB1-triggered SERA6 autoproteolytic processing. Fully mature SERA6 is an active proteolytic enzyme, demonstrated using a synthetic peptide substrate based on a predicted autocatalytic cleavage site. Small-molecule inhibitors of SERA6 autoprocessing block egress of asexual blood-stage parasites. 2026 PbSERA3 (the P. berghei ortholog of PfSERA6) is essential for hepatic merozoite egress: conditional mutant sporozoites develop normally in hepatocytes but fail to initiate blood-stage infection. PbSERA3 localizes to the surface of ookinetes and sporozoites under non-permeabilized conditions, indicating an extracellular location in those stages, and undergoes SUB1-mediated processing yielding two cleavage products in both blood and mosquito stages. Yeast-based Flp/FRT conditional mutagenesis, immunofluorescence under non-permeabilized conditions (surface localization), Western blot of processing products mBio Medium 41616267
