COX1 (PBANKA_MIT01800) is the mitochondrially encoded catalytic core subunit of cytochrome c oxidase in Plasmodium, expressed from the 6-kb extrachromosomal mitochondrial element during erythrocytic stages (PMID:1701017). In P. yoelii, COX1 is transcribed as a discrete 1.4 kb RNA whose 5' end carries a 12-nucleotide sequence (5'-TATTTTTGTTT-3') shared with the cytochrome b transcript and potentially functioning as a promoter or RNA processing signal, with a stem-loop at the end of the open reading frame serving as a putative transcription termination signal (PMID:1701017). Biochemically, in P. berghei COX1 is a subunit of an oligomeric cytochrome c oxidase complex of ~670 kDa minimal molecular weight that exhibits low catalytic efficiency (kcat/Km = 5.9 × 10⁻⁵ M⁻¹·min⁻¹) toward reduced cytochrome c (PMID:8397100). Beyond its role as the catalytic core of this complex and its transcript architecture, no direct functional characterization of the COX1 protein itself has been established in the available corpus; the P. falciparum copper assembly factors Cox11 and Cox19 are only predicted, by analogy to yeast, to deliver copper to the CuB site within COX1, without demonstrated interaction with COX1 (PMID:35672733, PMID:37348640).

Mechanistic history

Synthesis pass · year-by-year structured walk · 4 steps
  1. 1990 Medium

Establishing where and how COX1 is encoded and expressed addressed whether the parasite's cytochrome c oxidase core is mitochondrially encoded and how its transcription is regulated.

Evidence Northern blotting, primer extension, and RNA sequencing of P. yoelii erythrocytic-stage RNA using 6-kb DNA probes

    PMID:1701017

    Open questions at the time
    • The 12-nucleotide 5' sequence is only a candidate promoter/processing signal, not functionally validated by mutagenesis
    • The stem-loop is a putative terminator, not confirmed to terminate transcription
    • Does not address the protein's assembly or catalytic function
  2. 1993 Medium

Purifying the intact enzyme addressed the physical form and catalytic competence of the cytochrome c oxidase complex housing COX1 in the parasite.

Evidence Differential centrifugation, Percoll gradient, affinity chromatography, gel filtration, SDS-PAGE, and activity assay of P. berghei erythrocytic mitochondrial cytochrome c oxidase

    PMID:8397100

    Open questions at the time
    • COX1 subunit was not individually resolved or validated within the complex
    • The parasite SDS-PAGE profile was indistinguishable from host enzyme, limiting subunit-level conclusions
    • Low catalytic efficiency was measured but its mechanistic basis was not determined
  3. 1993 Low

Comparative sequence analysis addressed whether the COX1 protein is structurally conserved relative to other species, distinguishing it from the divergent cytochrome b.

Evidence DNA sequencing and comparative structural analysis of the P. falciparum 6-kb mitochondrial element

    PMID:8459834

    Open questions at the time
    • Structural inference is computational only, with no biochemical or functional validation of the COX1 protein
    • Predicted conservation does not establish activity or assembly in the parasite context
  4. 2023 Low

Characterizing copper chaperones PfCox11 and PfCox19 addressed how copper might be delivered to the CuB catalytic site of COX1 during enzyme maturation.

Evidence Recombinant expression and copper-binding assays with site-directed mutagenesis of PfCox11 (Cys157 critical) and Cx9C-motif copper binding by PfCox19 in P. falciparum

    PMID:35672733 PMID:37348640

    Open questions at the time
    • Copper delivery to the COX1 CuB site is an inference from yeast analogy; no direct interaction with COX1 was demonstrated
    • Experiments characterize the chaperones, not COX1 itself
    • The order and mechanism of chaperone-to-COX1 copper transfer are undefined

Open questions

Synthesis pass · forward-looking unresolved questions
  • Direct functional and structural characterization of the COX1 protein itself — its assembly, catalytic residues, and physical interaction with copper chaperones — remains unresolved.
  • No direct demonstration of COX1 interaction with Cox11 or Cox19
  • No structural model of the parasite COX1 subunit or its CuB site
  • No mutagenesis of COX1 to test catalytic or assembly roles

1990 In P. yoelii, the mitochondrial 6-kb extrachromosomal DNA encodes COX1 (cytochrome c oxidase subunit I); a discrete 1.4 kb RNA transcript encoding COX1 was identified during erythrocytic stages. Primer extension and RNA sequencing mapped a 5' end bearing the 12-nucleotide sequence 5'-TATTTTTGTTT-3', shared with the cytochrome b transcript, which may function as a promoter or RNA processing signal. A stem-loop structure at the end of the COX1 open reading frame was identified as a putative transcription termination signal. 1993 Cytochrome c oxidase was partially purified (~560-fold, 3.6% yield) from P. berghei erythrocytic-stage mitochondria by differential centrifugation, Percoll gradient, detergent solubilization, and reduced cytochrome c-agarose affinity chromatography. The native enzyme was an oligomeric complex with minimal molecular weight of ~670 kDa by gel filtration. The enzyme had low catalytic efficiency (kcat/Km = 5.9 × 10⁻⁵ M⁻¹·min⁻¹) toward reduced cytochrome c. SDS-PAGE profile of the parasite enzyme was not detectably different from that of host liver cytochrome c oxidase. Differential centrifugation, Percoll density gradient, affinity chromatography, FPLC gel filtration, SDS-PAGE, enzymatic activity assay with reduced cytochrome c Experimental parasitology Medium 8397100
1993 Sequence analysis of the P. falciparum 6-kb mitochondrial element predicted the structure of COX1 (cytochrome c oxidase subunit 1); the overall structure was found to be similar to COX1 from other species, in contrast to cytochrome b which showed striking divergence. 2022 In P. falciparum, the copper chaperone PfCox11 (assembly factor) binds copper via cysteine residues (Cys157 critical; Cys60 not required) and is predicted to deliver copper to the CuB site of the COX1 subunit of cytochrome c oxidase, based on functional analogy with yeast Cox11. Recombinant protein expression, affinity purification, copper binding assays (BCA release assay, atomic absorption spectroscopy, bacterial growth inhibition assay, ascorbate oxidation inhibition, thermal shift assay), site-directed mutagenesis of copper-binding cysteines Malaria journal Low 35672733
2023 In P. falciparum, PfCox19 binds copper via Cx9C motifs and is predicted to associate with PfCox11 to deliver copper to the CuB site of the COX1 subunit of cytochrome c oxidase, based on functional analogy with yeast Cox19. Recombinant expression, affinity purification, copper binding assays (BCA release assay, bacterial growth inhibition assay, ascorbate oxidation inhibition, differential scanning fluorimetry) Experimental parasitology Low 37348640
