Synthesis pass · prose summary of the discoveries below

CHT1 (PBANKA_0800500, PbCHT1) encodes the sole chitinase expressed by P. berghei ookinetes and functions in the parasite's traversal of the mosquito midgut during transmission (PMID:2011589, PMID:11349074). In P. berghei, chitinase activity is developmentally restricted, appearing only after zygote formation and coinciding with ookinete maturation, and it digests the chitin-containing peritrophic matrix that the ookinete must cross (PMID:2011589). Targeted disruption of PbCHT1 reduces mosquito infectivity by up to 90% and eliminates all residual ookinete-derived chitinase activity, confirming that this single gene accounts for ookinete chitinase and is required for efficient midgut invasion; the reduction seen even in feeds preceding peritrophic matrix formation indicates a role beyond matrix penetration, and the P. berghei enzyme is notably insensitive to allosamidin (PMID:11349074). The enzyme is secreted as an inactive prochitinase that is activated in the midgut by a trypsin-like protease, a step upstream and essential for transmission (PMID:8483942, PMID:8641775). Across Plasmodium orthologs, CHT1 is stored in ookinete micronemes and secreted apically, co-localizing with partners such as WARP and SOAP within a high-molecular-weight secreted complex (PMID:11035760, PMID:19206106, PMID:33489941), and structure-function work on the PfCHT1 ortholog assigns an intramolecular disulfide pair (cys-220/cys-230) to catalytic integrity and a surface cysteine (cys-203) to intermolecular complex assembly (PMID:35481637). Much mechanistic detail for CHT1 derives from orthologs (P. gallinaceum, P. falciparum); notably, chimeric P. berghei ookinetes expressing PfCHT1 produced only monomer, indicating that HMW complex formation depends on species-specific partners not present in P. berghei (PMID:33489941).

1991 P. berghei ookinetes produce a chitinase (CHT1 ortholog) that is absent until 15 hours after zygote formation and begins appearing in culture supernatant at 20 hours, coinciding with ookinete maturation; this enzyme digests the chitin-containing peritrophic matrix, supporting its role in peritrophic matrix penetration during midgut invasion. Native PAGE with chitinase activity staining, culture supernatant assay using 4-methylumbelliferyl chitotriose substrate, timing correlated with ookinete maturation Proceedings of the National Academy of Sciences of the United States of America Medium 2011589
1993 Chitinase activity secreted by Plasmodium berghei ookinetes is activated by a diisopropyl fluorophosphate-sensitive trypsin-like protease from the mosquito midgut (and by endoproteinase Lys-C); inhibition of chitinase with allosamidin completely blocks oocyst development in vivo, establishing that chitinase activity is essential for parasite transmission. In vivo allosamidin inhibition assay measuring oocyst formation; protease activation assay with midgut extracts and endoproteinase Lys-C; reversal with exogenous chitinase Proceedings of the National Academy of Sciences of the United States of America High 8483942
2001 Targeted gene knockout of PbCHT1 (PBANKA_0800500) in P. berghei reduced parasite infectivity in Anopheles stephensi mosquitoes by up to 90%; PbCHT1 null mutants had no residual ookinete-derived chitinase activity in vitro, indicating P. berghei ookinetes express only this single chitinase gene. Reduced infectivity was also seen in ookinete feeds before peritrophic matrix formation, suggesting PbCHT1 plays an additional role beyond peritrophic matrix disruption. PbCHT1 chitinase activity was insensitive to allosamidin inhibition, distinguishing it from human and avian malaria parasite chitinases. 2000 Plasmodium chitinase (PgCHT1 in P. gallinaceum; CHT1 orthologs) is concentrated at the apical end of ookinetes, is present within ookinete micronemes, and subsequently localizes to the electron-dense area of the apical complex, defining a micronemal secretory pathway for extracellular chitinase secretion. 2009 Chitinase (CHT1 ortholog) was identified as one of the most abundant proteins in isolated P. berghei ookinete micronemes by gel LC-MS/MS and MudPIT proteomics, confirming its micronemal localization at the protein level. 2004 Chitinase (CHT1 orthologs from P. gallinaceum: PgCHT1 soluble; CTRP and WARP membrane-associated) co-localize to the same micronemal population as shown by immunogold double-labeling EM; within the microneme, PgCHT1 and WARP are diffuse in the lumenal space whereas CTRP is peripherally associated with the microneme membrane, establishing differential sub-micronemal distribution. 2021 P. falciparum PfCHT1 (short-form chitinase, ortholog of PbCHT1/PBANKA_0800500) is a covalently linked component of a high-molecular-weight (>1,300 kDa) chitinase-containing complex secreted by ookinetes; mass spectrometry identified WARP, ookinete surface enolase, and SOAP as components of this complex. Chimeric P. berghei ookinetes engineered to express PfCHT1 in place of PbCHT1 produced only monomeric PfCHT1 without forming the HMW complex, indicating that HMW complex formation requires P. falciparum-specific or P. gallinaceum-specific binding partners absent in P. berghei. 1996 Mosquito midgut trypsin activates the Plasmodium prochitinase (CHT1 precursor); trypsin activity and chitinase-activating activity co-migrated on gels, and trypsin-specific inhibitors blocked both prochitinase activation and peritrophic matrix penetration. Anti-trypsin antibodies blocked infectivity in a manner reversible by exogenous chitinase, demonstrating that trypsin acts upstream of chitinase activation in the transmission pathway. 1D gel co-migration assay, trypsin inhibitor feeding experiments, anti-trypsin antibody membrane-feeding assay, reversal with exogenous chitinase Infection and immunity High 8641775
2021 AlphaFold structural modeling of short-form chitinases (including PfCHT1, ortholog of PbCHT1) predicted a three-dimensional structure revealing how short chitinase, WARP, and SOAP may interact at the atomic level to form a hetero-multimeric ookinete-secreted peritrophic matrix invasion complex; conserved residues across Plasmodium species were identified in this complex interface. 2022 Structure-function analysis of the three cysteines of PfCHT1 (short-form chitinase, ortholog of PbCHT1) showed that cys-220 and cys-230 form intramolecular disulfide bonds essential for proper folding and enzymatic activity (their mutation abolished chitinolytic activity and chitin binding), whereas cys-203 is surface-exposed, dispensable for catalytic and chitin-binding functions, and predicted to mediate intermolecular protein-protein interactions in the HMW complex.
