PBANKA_0304900 (SERA3) encodes a papain-domain-containing serine repeat antigen that is expressed, presented at the parasite surface, and proteolytically processed across multiple Plasmodium life-cycle stages, where it functions in host-cell disruption and merozoite egress (PMID:18419771, PMID:41616267). In P. berghei, SERA3 is transcribed and its protein is processed during late liver stages, and it leaks into the hepatocyte cytoplasm during merozoite development, positioning it to contribute to host cell death (PMID:18419771). Conditional mutagenesis established that P. berghei SERA3 is dispensable for hepatocyte invasion and late liver-stage development but is essential for hepatic merozoite egress and the transition to blood-stage infection (PMID:41616267); it also localizes to the surface of ookinetes and sporozoites and is processed at these stages, with SUB1 (subtilisin-like protease) identified as the protease responsible for its maturation (PMID:41616267). Consistent with a non-essential blood-stage role, SERA3 is dispensable for erythrocytic growth in P. falciparum, in contrast to the essential SERA4–6 members of the same locus (PMID:12228245), and is expressed in trophozoite and schizont stages within the coordinately regulated chromosome 2 SERA cluster (PMID:7891737, PMID:12244052). Beyond its processing by SUB1 and its role in hepatic egress, the biochemical activity of the SERA3 papain domain and its downstream targets during host cell disruption have not been characterized in the available corpus.
